摘  要:目的分析Smad7与NF-κBp65在原发性肝细胞性肝癌(HCC)中的表达、临床意义及两者之间的关系。方法选择2005年12月~2010年12月顺德第一人民医院外科手术切除送检至病理科的腊块标本120例为研究对象,其中正常肝组织10例、肝炎组织30例、肝硬化组织30例、HCC组织50例。采用免疫组化S-P法和石蜡切片原位杂交法检测各肝组织中Smad7 mRNA、NF-κBp65 mRNA及其蛋白的表达情况。结果 Smad7蛋白、NF-κBp65蛋白、Smad7 mRNA、NF-κBp65 mRNA在HCC组织中的阳性表达率均明显高于肝硬化、肝炎和正常肝组织(P<0.01)。肝硬化组织中Smad7 mRNA、Smad7蛋白的阳性表达率明显高于正常肝组织(χ2=4.043、4.848,均P<0.05)。HCC中NF-κBp65蛋白的表达与肿瘤直径及Smad7蛋白的表达有关(χ2=5.592、4.578,均P<0.05)。结论 Smad7和NF-κBp65异常表达可能共同参与了HCC的演进。Objective To study the expression of Smad7 and NF-κBp65 in HCC and their relationship.Methods 120 cases of specimen from patients carried surgical resection in the First People′s Hospital of Shunde of Southern Medical University from December 2005 to December 2010 were selected as the research object,including 10 cases of normal hepatic tissues,30 case of cirrhosis,30 cases of hepatitis and 50 cases of HCC.Immunohistochemistry and in situ hybridization were used to detect the changes of Smad7 mRNA and NF-κBp65 mRNA and their proteins expressions in each kinds of hepatic tissues.Results The positive expressions rate of Smad7 mRNA and NF-κBp65 mRNA and their protein in HCC tissues were significantly higher than those in the cirrhosis tissues,hepatitis tissues and normal hepatic tissues,the differences were statistically significant(P〈0.01).The expression of Smad7 mRNA and proteins in cirrhosis tissues were higher than those in normal tissues(χ2=4.043,4.848,all P〈0.05).The expression of NF-κBp65 protein in hepatocellular carcinoma was related to tumor size and the expression of Smad7 protein(χ2=5.592,4.578,all P〈0.05).Conclusion The abnormal expression of Smad7 and NF-κBp65 may be related to the evolution of HCC.
