摘　　要：目的探讨神经生长因子(NGF)诱导PC12细胞分化为神经元样细胞时MAP2的表达变化情况.方法细胞培养6 d后应用共聚焦显微镜观察MAP2表达情况;Western Blot法检测诱导不同时间点MAP2的表达情况.结果免疫组化结果显示:MAP2表达呈阳性;免疫印迹结果显示:MAP2蛋白表达在不同时间点呈动态变化,即随诱导时间延长呈递增趋势.结论 NGF成功诱导PC12细胞分化为神经样细胞,为缺血性脑卒中的体外细胞研究提供细胞参考.Objective To investigate the expression change of MAP2 in the differentiation of PC12 cells into neuron like cells induced by nerve growth factor（NGF）.Method The cells cultured for 6 days out of training board,discard the original culture liquid,immersion,fixed,proteinase K digestion,clamping incubated for half an hour,with a resistance,overnight,dip,plus two anti dropping half hour incubation under confocal microscopy.The expression of MAP2 was detected with Western Blot induced by MAP2 at different time points.Results Immunohistochemistry showed that MAP2 expression was positive.Western blot showed that the expression of MAP2 protein changed dynamically at different time points.Conclusion NGF successfully induces differentiation of PC12 cells into neuron like cells,providing the cellular basis for in vitro studies of ischemic stroke.
