摘 要:目的:探讨应用低功率超声放大卵巢癌肿瘤标志物提高卵巢癌早期检出率的可行性。方法:用自制的超声信号发生装置,设定超声信号频率为1 MHz,超声功率为0.1~0.3 W/cm^2,时间为1、2、3、4、5分钟,对人卵巢癌细胞株SKOV3行超声辐射,检测培养基中的肿瘤标志物;以功率0.1~0.3 W/cm^2和超声辐射时间5、10、15分钟对裸鼠卵巢癌瘤体进行声辐射后检测血肿瘤标志物,对标志物的放大效应进行分析。结果:在细胞水平,超声辐射时间一定的条件下,CA125浓度随功率增大而增加,且差异有统计学意义(P<0.05)。0.2 W/cm^2和0.3 W/cm^2超声条件下,CA125浓度随照射时间延长而增加,不同时间组间差异均有统计学意义(P<0.05)。相同的超声辐射时间条件下,CA199浓度随功率的提高而增加(P<0.05)。经超声辐射后,各组裸鼠血清CA_(125)、CA_(199)浓度较对照组均显著上升(P<0.05),不同功率组间差异有统计学意义(P<0.05)。结论:低功率超声可在细胞和瘤体使卵巢癌肿瘤标志物CA_(125)和CA_(199)浓度增加。Objective:To explore the feasibility of provide a feasible method in early detection of ovarian cancer by raising the TMs concentration of ovarian cancer after low-intensity ultrasound exposure. Methods:Sonication to epithelial ovarian cancer cells (SKOV3) by using a self made 1 MHz ultrasound generator (0. 1 - 0.3 W/cm2,1 -5 min), and the TMs concentration in culture medium were detected. Based on the model of nude mouse xenograft,experiments were carried out to verify the feasibility of ultrasounic amplification of TMs, by using the ultrasound generator(0. 1 -0.3 W/cm2, 5 rain, 10 rain, 15 min). Results: In the cell level, with the same ultrasonic irradiation time, both CA125 concentrations were elevated when the power increased ( P 〈 0.05). With the 0.2 W/cm2 and 0.3 W/cm2 ultrosound,the concentration of CA125 increased followed by the increase of ultrasonic time( P 〈 0.05). With the same ultrasonic time,the CA199 concentrations were elevated when the power increased (P 〈0.05). After treated by sonication, serum CA125 and CA199 concentration of nude mice were significantly higher than those of control group ( P 〈 0.05), and there were significant difference among the different power groups ( P 〈 0.05). Conclusions: Ovarian cancer TM CA 125 and CA 199 increase by low-intensity ultrasounic irradiation in vivo and vitro.
