摘 要:目的:探索鼻咽癌患者在放射治疗过程中鼻咽部细菌群落结构的动态变化情况。方法:取7例鼻咽癌患者在放疗过程中不同时点的鼻咽拭子样本,利用T-RFLP技术对其鼻咽部菌群结构进行分析。结果:在放疗不同时点的鼻咽拭子样本中共获取115种末端限制性片段(T-RFs),其中以放疗前的数目最多(91种),放疗结束时最少(49种),片段62bp、65bp、77bp、120bp、146bp、253bp、333bp、398bp、400bp、469bp所代表的菌群贯穿于整个放疗周期中。鼻咽部菌群结构随放疗时间变化而改变,至放疗结束时T-RFs数量下降46%,香农指数(H)下降5%,均匀度指数(E)增高5%,辛普森指数(D)变化趋势不明显。结论:放疗可影响鼻咽癌患者鼻咽部菌群的生长,致群落中的T-RFs数目减少,优势细菌分布发生变化,菌群结构多样化程度下降。Objective: To reveal the dynamic changes of nasopharyngeal microbiome during radiotherapy in nasopharyngeal carcinoma (NPC) patients. Methods: The nasopharyngeal swabs samples were collected from 7 NPC patients during radiotherapy at different time points. The structural changes of microbiome in nasopharynx were analyzed by terminal restriction fragment length polymorphism technology (T-RFLP). Results: Among 115 terminal restriction fragments (T-RFs) obtained in total, 91 T-RFs were yield from the pharyngeal swabs before treatment and 49 T-RFs at the end of radiotherapy. The bacterial T-RFs with 62 bp, 65 bp, 77 bp, 120 bp, 146 bp, 253 bp, 333 bp, 398 bp, 400 bp and 469 bp existed through the course of radiotherapy. The percentage of T-RFs was decreased to 46% at the end of radiotherapy, the Shannon index was decreased to 5 %, and the evenness index was increased to 5%.
