摘  要:目的:探讨细胞周期素依赖激酶样蛋白1(CDKL1)对胃癌细胞系SGC7901迁移及侵袭的作用。方法:将胃癌细胞系SGC7901分为实验组及对照组,采用慢病毒转染siRNA至胃癌细胞构成实验组,转染无意义序列作为对照组。通过细胞划痕实验检测胃癌SGC7901细胞的迁移能力;Transwell小室实验检测其侵袭能力;Western blot检测CDKL1低表达后对胃癌细胞中AEG-1和MMP-9蛋白表达的影响。结果:与对照组相比,CDKL1低表达后实验组细胞迁移及侵袭能力明显降低(P<0.05),实验组胃癌细胞中AEG-1和MMP-9蛋白的表达水平明显下降(P<0.05)。结论:特异性下调CDKL1基因可抑制胃癌细胞的迁移及侵袭能力,表明CDKL1基因可促进胃癌细胞的侵袭以及转移,并且与AEG-1和MMP-9密切相关。Objective: To investigate the effects of cyclin-dependent kinase-like 1(CDKL1) on migration and invasion of gastric cancer cells.Methods: Gastric cancer cells SGC7901 were divided into siRNA interference groups and control group.Cell scratch method and Transwell experiment were used to detect the ability of migration and invasion of SGC7901 cells in each group.Astrocyte elevated gene-1(AEG-1) and matrix metalloproteinase-9(MMP-9) protein were determined by Western blot respectively.Results: Compared with the control group,the abilities of migration and invasion of SGC7901 cells in the interference group were decreased remarkably(p〈0.05).The AEG-1 and MMP-9 protein expression of SGC7 9 0 1 cells in interference group decreased remarkably(p〈0.05).Conclusion: The CDKL1 can promote migration and invasion of gastric cancer cells,which is related to AEG-1 and MMP-9 protein probably.
