摘 要:目的探讨DACH2基因在膀胱尿路上皮癌组织中的表达情况,并分析其与膀胱癌临床病理特征及复发的关系。方法应用免疫组化技术(SP法)和实时定量聚合酶链反应(q RT-PCR)法检测47例膀胱尿路上皮癌组织及11例癌旁正常膀胱黏膜组织中DACH2的表达,并结合临床病理资料进行分析。结果免疫组化结果显示DACH2蛋白在肿瘤组表达高于正常膀胱黏膜组(P<0.05);随分级、分期增高,DACH2蛋白组间表达差异有统计学意义(P<0.05)。q RT-PCR显示DACH2的m RNA表达在肿瘤组(2-△△Ct 18.25)高于正常膀胱黏膜(2-△△Ct 1.00)(P<0.05);复发组(2-△△Ct 19.70)高于原发组(2-△△Ct 7.57)(P<0.05);随膀胱癌的分级、分期增高,DACH2的m RNA组间表达差异有统计学意义(<0.05)。结论 DACH2高表达与膀胱尿路上皮癌的恶性侵袭性明显相关,检测DACH2有助于膀胱癌的分期和分级。Objective To explore the expression of dachshund homolog 2 gene (DACH2) in bladder urothelial cancer and analyze its clinical significance and relationship with tumor biologic characteristics. MethodsThe expressions of DACH2 in 47 specimens of bladder cancer and 11 specimens of normal bladder tissues were detected by immunohistochemical staining and quantitative RT-PCR, and its clinical significance was then analyzed. Results The expression of DACH2 protein was higher in bladder cancer than that in the adjacent normal tissues (P〈0.05). Expression of DACH2 protein was higher in muscle invasive bladder cancers than in non-muscle invasive bladder cancer (P〈0.05). Quantitative RT-PCR analyses of DACH2 expression showed that there was a significant difference between bladder cancer and the adjacent normal tissues (2-△△α value, 18.25 vs 1.00,P〈0.05). The expression of DACH2 gene was significantly increased in recurrent bladder cancer tissues compared with the primary cancer group (2-△△αvalue, 19.70 vs 7.57, P〈0.05). Expression of DACH2 was elevated in higher stage and grade bladder cancers than that in lower stage and grade bladder cancer (P〈0.05). Conclusion DACH2 may play a role in the development of bladder urothelial cancer. The determination of DACH2 expression is potentially valuable in diagnosis and prognostic evaluation for bladder urothelial cancers.
