“通督治郁”针法对脑卒中后抑郁模型大鼠体质量、行为学及海马部位环磷腺苷反应元件结合蛋白水平的影响

摘　　要：目的观察"通督治郁"针法对脑卒中后抑郁(PSD)模型大鼠的体质量、行为学及海马部位环磷腺苷反应元件结合蛋白(CREB)水平的影响。方法将SD大鼠随机分为正常组、模型组、治疗组,除正常组外,模型组和治疗组大鼠采用大脑中动脉线栓阻塞法联合慢性不可预知性温和应激处理方法制备PSD模型。治疗组采用"通督治郁"针法治疗。记录不同阶段大鼠体质量,通过敞箱实验观察大鼠的行为学变化,通过免疫组化法测定海马部位CREB的表达。结果 PSD大鼠模型制备成功后,模型组大鼠体质量明显减轻,敞箱实验水平运动和垂直运动明显减少,海马部位CREB表达明显降低(P<0.01);通过"通督治郁"针法干预4周后,与模型组比较,治疗组大鼠体质量明显增加,敞箱实验水平运动和垂直运动显著增加,海马部位CREB表达明显升高(P<0.05或P<0.01)。结论 "通督治郁"针法可促使PSD模型大鼠体质量的增加,提高大鼠行为学评分及海马部位CREB的表达水平。Objective To observe the effect of Tongdu Zhiyu acupuncture therapy（ techniques of needling in Ren and Du meridians for adjustment and smoothing） on body mass,behavioral score and c AMP-response element binding（ CREB） protein expression in hippocampus of model rats with post-stroke depression（ PSD）. Methods Rats were randomized into normal group,model group and treatment group. Except the normal group,other rats were made into PSD models by occlusion of middle cerebral artery（ MCAO） and chronic unpredictable mild stress processing. Then the treatment group was intervened with Tongdu Zhiyu acupuncture therapy. Body mass were recorded at different stages,behavior changes were observed by open field test and expression of CREB was measured by immunocytoche-mistry. Results After modeling,rats’ body mass lost,horizontal and vertical movements decreased and expression level of CREB lowered obviously（ P 〈 0.01）. On 4thweek after acupuncture,compared with model group,the treatment group had body weight gained,horizontal and vertical movements increased as well as CREB level elevated remarkably（ P〈0.05 or P〈0.01）. Conclusion Tongdu Zhiyu acupuncture therapy is a good therapy to put on weight,improve behavior score and raise the level of CREB expression in PSD rats.
