MTHFR和CBS甲基化水平与哈萨克族食管癌的相关性研究

摘　　要：目的探讨叶酸代谢相关基因亚甲基四氢叶酸还原酶基因(MTHFR)和胱硫醚-β-合酶基因(CBS)DNA甲基化水平与哈萨克族食管癌的关系。方法选择2010-2015年新疆哈萨克族食管癌患者192例(病例组),非食管癌对照200例(对照组),采用甲基化特异性聚合酶链反应(MSP)技术检测病例组术前外周血和对照组外周血中MTHFR和CBS基因启动子区甲基化水平。结果病例组外周血中MTHFR基因完全甲基化、部分甲基化和未甲基化的检出率分别为25.5%(49/192)、31.8%(61/192)和42.7%(82/192),对照组分别为11.0%(22/200)、13.0%(26/200)、76.0%(152/200)。病例组MTHFR基因甲基化检出率明显高于对照组,差异有统计学意义(χ2=45.144,P<0.05)。病例组CBS基因完全甲基化、部分甲基化和未甲基化的检出率分别为7.8%(15/192)、9.9%(19/192)和82.3%(158/192),对照组分别为0.0%(0/200)、16.0%(32/200)、84.0%(168/200),病例组CBS基因甲基化率高于对照组,差异有统计学意义(χ2=18.465,P<0.05)。结论叶酸代谢相关基因MTHFR和CBS甲基化水平与哈萨克族食管癌的发生密切相关。Objective To investigate the relationship between DNA methylation level of folate metabolism related genes MTHFRand CBSwith esophageal cancer in kazakh.Methods 182 kazakh patients of esophageal cancer and 200 kazakh patients of non-esophageal cancer in Xinjiang were collected from 2010to2015.The DNA methylations of MTHFRand CBSgene were detected by MSP.Results The detection rate of complete methylation,partial methylation and unmethylation of MTHFR gene in the case group were 25.5%（49/192）,31.8%（61/192）and 42.7%（82/192）,while there were 11.0%（22/200）,13.0%（26/200）,76.0%（152/200）in the control group.The methylation rate of MTHFRin the case group was significantly higher than that in the control group,and the difference was statistically significant（χ^2=45.144,P〈0.05）.The detection rate of complete methylation,partial methylation and unmethylation ofCBSgene in the case group were 7.8%（15/192）,9.9%（19/192）,82.3%（158/192）,while there were0.0%（0/200）,16.0%（32/200）and 84.0%（168/200）in the control group.The methylation rate of CBS in the case group was higher than that in the control group,and the difference was statistically significant（χ^2=18.465,P〈0.05）.Conclusion The methylation levels of folate metabolism related genes of MTHFRgene and CBSgene are closely related to esophageal cancer in Kazakh.
