摘  要:LPBG08是本研究室培育的一个特大粒水稻种质,其平均粒长达15 mm,平均粒宽达4.2 mm,平均千粒重达66 g。为了解析该特异种质粒形性状形成的遗传机理,本研究以其为供试材料,利用目的基因扩增和测序手段,对2个重要粒长基因(GS3,qGL3/GL3.1)和4个重要粒宽基因(GW2,GW8,GS5和GW5)进行检测。检测发现,6个粒形相关基因中有5个在LPBG08中发生了正调控突变。其中,GS3和qGL3/GL3.1 2个粒长基因全都发生突变,4个控制粒宽基因中的GW2、GW8和GW5等3个发生突变。研究结果表明,LPBG08的特大粒粒形性状的形成可能是由于多个正向调控突变的粒形相关基因聚合所引起的累加效应。LPBG08 is a special germplasm with extra large grains bred by our lab. The average grain length and width of LPBG08 is about 15 mm and 4.2 mm, respectively, and average 1 000-grain weight is about 66 g. In order to analyze the genetic mechanism of the formation of grain shape trait in the special germplasm, in this research,with LPBG08 as research material, we detected 2 important grain length genes, GS3 and qGL3/GL3.1, and 4important grain width genes, GW2,%GW8, GS5 and GW5 by amplifying and sequencing target genes. Based on the detection results, it was found that positive-regulation mutations had occurred on five out of the six genes in LPBG08. Among them, both of the two grain length genes, GS3 and qGL3/GL3.1, and three grain width genes,GW2, GW8 and GW5, were mutant. The research results suggested that the formation of extra large grain trait in LPBG08 might be the result of additive effects from the pyramiding of multiple grain-shape-related genes that positively regulate mutation.
