摘  要:【目的】通过研究不同体积分数NaCl胁迫下不同马铃薯试管苗材料生长生理指标及离子分布的影响,为马铃薯耐盐性育种提供理论依据.【方法】以引自国际马铃薯中心的4个马铃薯试管苗种质资源CIP391047.34(简称A)、CIP385499.11(简称B)、CIP394611.112(简称C)和CIP391919.3(简称D)为材料,采用组织培养技术研究不同体积分数(0、0.3%、0.6%、0.9%、1.2%)NaCl处理对马铃薯试管苗生长生理指标及Na^+和K^+含量的影响,比较其耐盐性.【结果】不同体积分数NaCl处理下,4个引进马铃薯试管苗材料的新叶数、根数、最长根长、茎叶鲜质量、根鲜质量和根冠比均较对照显著下降,其中0.3%的NaCl逆境对试管苗根数影响较大,4个材料的处理与对照间均差异显著,且B材料下降幅度最小、D材料下降幅度最大;0.9%NaCl逆境下试管苗根或叶的形成及生长严重抑制,1.2%NaCl逆境下试管苗生长基本停滞,且C和D材料没有根的发生.盐逆境下,4个材料试管苗的脯氨酸含量、丙二醛含量及Na^+/K^+的变化趋势一致,均随盐体积分数的升高而上升,0.9%NaCl处理下,D材料的脯氨酸含量、丙二醛含量及Na^+/K^+(茎叶)分别是B材料的2.67、1.80和1.87倍.【结论】本研究选用的种质材料可耐受盐害体积分数为≤0.6%,B材料最耐盐、D材料对盐最敏感,脯氨酸可作为胁迫伤害的评价指标.[Objective] The study was to analyze the influence of different concentrations of NaCl on the growth, physiological indexes and ion distribution of potato plantlets to provide theoretical basis for the po- tato salt tolerance breeding. [Method] Taking four potato lines from International Potato Centre (CIP391047.34,termed as A, CIP385499. 11, termed as B, CIP394611. 112,termed as C, CIP391919. 3, termed as D) as the material, the Growth and physiological indexes, Na+ and K+ content of were investiga- ted to assess salt tolerance under different NaC1 concentrations (0,0. 3%,0. 6%,0. 9%, 1.2 %). [Result] The number of young leaves, root number, maximum root length, fresh weight of stem leaf and root, root- shoot ratio of four introduced lines declined significantly compared with the control under different NaCl treatments. Root number of all lines was dramatically impacted at 0. 3 % NaCl concentration, the decreasing range of root number in the line B was the minimum while in the line D was the maximum. The formation and growth of roots and leaves were severely inhibited at 0. 9% NaCl concentration. The growth of plant- lets nearly stopped and there was no root in the lines C and D at 1.2 % NaCl concentration. The content of proline, malondialdehyde and ratio of Na+ to K+ of 4 lines increased with the rise of salt concentration. The content of proline,malondialdehyde and ratio of Na+ to K+ of line D were as 2. 67,1.80 and 1.87 times as that of the line B, respectively. [Conclusion] The threshold of salt tolerance of 4 lines was 0. 6G. The line B is salt tolerant and line D is salt sensitive. The proline content can be used as an evaluation index under salt stress.
