摘  要:目的本研究通过分析比较遗传性癫痫大鼠(Tremor,TRM)与正常大鼠(Wistar)海马中BDNF蛋白表达与定位差异,进一步阐明癫痫发病机制。方法以正常Wistar大鼠为对照组,应用PCR技术,筛选阳性自发性癫痫大鼠TRM作为实验组;通过Western blot技术分别对TRM及Wistar大鼠海马中BDNF蛋白进行半定量检测;免疫组织化学技术分别测定TRM及Wistar大鼠海马中BDNF蛋白的定位表达变化。结果与Wistar大鼠相比,TRM大鼠的海马中BDNF蛋白表达无明显变化;BDNF蛋白在海马DG区和CA1区定位表达减少。结论与Wistar大鼠相比,TRM大鼠海马DG区和CA1区BDNF蛋白定位表达减少,表明癫痫的发作可能与BDNF蛋白在海马区定位的异常变化有关。Objective To detect the change of brain derived neurotrophic factor in the hippocampus of genetic epileptic rats (Tremor, TRM) and Wistar rats, in order to further clarify the pathogenesis of epilepsy. Methods Normal rats were applied as control group, and TRM rats were identified by PCR technique as model group. The expression of BDNF protein was determined by Western blot ; the localization of BDNF in hippocampus of TRM rats and Wistar rats was detected by immunohistochemistry. Results The BDNF expression was not changed in the hippocampus of TRM rats compared with that of control group ; The BDNF localization was down-regulated in the hippocampal DG and CA1 regions of TRM rats compared with that of control group. Conclusion Down-regulation of BDNF protein localization in the hippocampal DG area and CA 1 area of TRM rats may be involved in the mechanism of epilepsy.
