摘 要:先在硅胶表面接枝偶氮活性基团,在此基础以去氧肾上腺素(Phenylephrine,PE)为替代模板分子、丙烯酰胺(Acrylamide,AM)为功能单体在其表面合成具有特定识别克伦特罗(Clenbuterol,CLB)分子印迹聚合物。计算机分子模拟确定了功能单体与模板分子最佳比例为3:1;对各步合成的硅胶进行了表征证明成功合成了聚合物;通过对比不同反应条件下所得聚合物对CLB吸附率筛选出了最佳聚合反应条件(65℃,12 h);对不同浓度梯度CLB标准液进行高效液相色谱分析,绘制出标准曲线,并以此分别测定CLB标准液低速流入所合成聚合物为填料的固相萃取柱的浓度差计算聚合物的吸附率;最后通过对比不加模板所合成聚合物和硅胶对CLB的吸附率,证明了所合成聚合物具有特异识别CLB能力。合成的聚合物可用于检测克伦特罗时的样品前处理,或作为传感元件进一步研发快速检测克仑特罗的传感器。Dummy molecularly imprinting polymers were grafted on silica bead using the phenylephrine(PE) as dummy template and acrylamide(AM) as functional monomers. The ratios of template molecular to functional monomer were determined by computational simulation. The succesful synthesis was confirmed by characterizing the silica bead after every step reaction. The optimum reaction condition with 65 ℃ and 12 h was obtained by comparing the results of different reaction conditions. The calibration curve line of CLB was obtained by analyzing the different concentrations of clenbuterol(CLB) standards solutions with HPLC and the adsorption of the prepared polymers was measured. The results indicated that the synthesized molecular imprinted polymers(MIPs) hold specific affinity to the target analyte of CLB, and it could be applied for rapid pretreatment of extraction of samples contaminated with CLB before determination or directly as sensing element in sensor for rapid detection of CLB in contaminated food sample.
