摘  要:[目的]探讨阿霉素(ADM)诱导人乳腺癌细胞MCF-7上皮-间质转化的作用.[方法]体外培养人乳腺癌细胞MCF-7,用不同质量浓度ADM处理后观察形态变化,并采用细胞免疫荧光染色法检测E-cadherin和vimentin蛋白水平.[结果]ADM可使MCF-7细胞变长变扁,明显增加MCF-7细胞的vimentin蛋白水平(P<0.05),降低E-cadherin蛋白水平(P<0.05).[结论]ADM可诱导人乳腺癌细胞MCF-7上皮-间质转化.OBJECTIVE To study the effects of adriamycin(ADM)on inducing epithelial-mesenchymal transition of human breast cancer cell line MCF-7.METHODS Human breast cancer cell line MCF-7 was cultured in vitro with different concentration of ADM,and the morphologic changes of the cells were observed.Immunofluorescence staining technique was used to detect the levels of E-cadherin and vimentin.RESULTSADM could make the MCF-7cell to be long and flat.ADM significantly increased the level of vimentin and decreased the level of E-cadherin in the MCF-7cells(P〈0.05).CONCLUSION ADM could induce epithelial-mesenchymal transition of human breast cancer cell line MCF-7.
