摘 要:目的研究亲环素J在肝癌发生中的作用。方法构建稳定表达CYPJ的肝细胞系,运用集落生成实验和裸鼠成瘤实验检测CYPJ是否促进肝癌的发生。结果集落形成实验中,转染CYPJ的L02细胞克隆数约是对照组的2倍(P﹤0.05);裸鼠成瘤实验中,CYPJ组肿瘤的重量约是对照组的4倍(P﹤0.05)。结论体外体内实验均证实CYPJ可促进肝细胞发生癌变,可为肝癌的治疗提供新的治疗靶点。Objective To investigate the role of cyclophilin J(CYPJ) in liver cancer oncogenesis. Method CYPJ plasmid was transfected into liver cells to establish a liver cell line expressing CYPJ stably, colony formation assay and tumor development assay in athymic nude mice were used to detect whether CYPJ could promote the occurrence of liver cancer. Result In colony formation assay, the colony number of cells transfected with CYPJ was about twice as many as the control group(P〈0.05). In the animal experiment, tumors originated from the CYPJ cells weighted three times more than the vector controls(P〈0.05). Conclusion CYPJ could promote liver tumor formation as shown in both in vitro and in vivo studies, which may be a potential therapeutic target for liver cancer treatment.
