摘 要:目的研究骨髓单个核细胞对脑梗死后脑源性神经营养因子(BDNF)及室管膜下区神经干细胞增殖的影响,探讨骨髓单个核细胞对脑梗死治疗作用的可能机制。方法成年雄性SD大鼠72只,体质量250-300g。梯度离心法分离骨髓单个核细胞,流式细胞术检测细胞纯度,将由模型动物股骨骨髓腔内分离出的单个核细胞经尾静脉途径移植到大鼠体内;线栓法制作脑梗死动物模型,分别于脑梗死后24h、72h通过Western blot技术检测脑部BDNF的表达;室管膜下区神经干细胞的增殖情况于脑梗死7d时通过BrdU/Nestin免疫荧光双染技术进行检测。结果经骨髓单个核细胞移植24h、72h时,BDNF表达的水平在脑梗死大鼠脑部明显增高,与模型组及溶剂组比较,差异有统计学意义(P〈0.01)。自体骨髓单个核细胞移植还促进了脑梗死7d时室管膜下区神经干细胞的增殖,骨髓单个核细胞移植组与模型组以及溶剂治疗组比较,差异亦具有统计学意义(P〈0.01)。结论自体骨髓单个核细胞移植可以显著增加脑梗死大鼠脑部BDNF的表达、并促进室管膜下区神经干细胞的动员。骨髓单个核细胞促进神经干细胞增殖的作用可能与骨髓单个核细胞的神经保护作用有关。Objective Our study aimed at investigating the effects of bone marrow mononuclear cells(BMMNCs)on brain-derived neurotrophic factors(BDNF)and on proliferation of neural stem cells in subventricular zone in rats with cerebral infarction,in order to explore the potential mechanisms of BMMNCs on cerebral infarction.Methods Totally 72 adult male Sprague-Dawley rats weighing 250 to 300g were used in this study.We performed thread occlusion technique to establish the model of cerebral infarction.Mononuclear cells(MNCs)isolated from femoral marrow cavity were separated by using densitygradient centrifugation method and the purity of selected cells was identified by flow cytometer.Then the separated MNCs were transplanted into rats via tail vein injection.The expression of BDNF in brain was detected by Western blotting method at24 hand 74hafter stroke and proliferation of neural stem cells in subventricular zone was tested by double BrdU/Nestin immu-nofluorescence staining on the seventh day after stroke.Results At 24 hand 72hafter auto-transplantation with bone marrow mononuclear cells,the expression of BDNF in transplantation group was obviously increased and showed statistical differences compared with model group and solvent group(all P〈0.05).Proliferation of neural stem cells in subventricular zone in the transplantation group was significantly improved,which presented statistical differences relative to both model group and solvent group(all P〈0.05).Conclusion BMMNCs can not only significantly increase the expression of BDNF but also effectively improve proliferation of neural stem cells in subventricular zone.The improvement of BMMNCs may be closely associated with the protective effect of BMMNCs.
