摘 要:目的 探讨血清miR系列基因表达水平与非小细胞肺癌(NSCLC)预后的相关性。方法分别收集2010年4月~2013年4月收治的250例NSCLC患者冰冻组织和血清样本,25例NSCLC及其癌旁组织,25例未经手术治疗的晚期NSCLC患者化疗前和3个疗程后的血清样本,200例未经任何抗癌治疗的NSCLC患者血清,作为NSCLC组;选取100例健康志愿者的血清样本,作为对照组。采用qRT-PCR方法检测NSCLC组和对照组中血清miR系列基因表达水平,判断血清miR系列基因表达水平与预后的关系。结果NSCLC组化疗3个疗程的患者的血清miR系列基因表达与化疗前存在明显差异(P<0.05);对照组的血清miR系列基因表达与化疗前患者存在明显差异(P<0.05),与化疗后指标比较无统计学意义(P>0.05)。结论血清miR系列基因表达水平在NSCLC患者明显升高,治疗后miR系列基因表达明显下降,血清miR系列基因有可能成为预测NSCLC预后的分子标志物。Objective To investigate the correlation between the expression level of serum miR gene and the prognosis of non-small cell lung cancer (NSCLC). Methods Frozen tissues and serum samples from 250 patients with NSCLC from April 2010 to April 2013 were collected. The serum samples of 25 cases of NSCLC and its adjacent tissues, 25 patients with advanced NSCLC without surgery before the chemotherapy and after 3 courses treatment, and 200 patients with NSCLC without any anticancer treatment were selected as the NSCLC group. Serum samples of 100 healthy volunteers were selected as the control group. The levels of serum miR gene expression in NSCLC group and control group were detected by qRT-PCR method, and the relationship between the expression level of serum miR gene and prognosis was judged. Results The expression of serum miR gene in the patients in NSCLC group after 3 courses of chemotherapy was significantly different from that before the chemotherapy(P〈0.05); the expression of serum miR gene in the control group was significantly different from that before chemotherapy(P〈0.05), and there was no significant difference in the patients after chemotherapy(P〉0.05). Conclusion Serum miR gene expression level is significantly increased in NSCLC patients. After the treatment, the expression of miR gene is decreased significantly. The serum miR gene may become a molecular marker for predicting the prognosis of NSCLC.
