摘 要:目的:优选苦豆草的酶法提取工艺,提高苦豆草生物碱的提取率。方法:以苦豆草生物碱为指标,通过单因素实验及正交试验优化酶质量浓度、pH、酶解温度、酶解时间等工艺参数。结果:最佳提取工艺条件为:酶解时间4h,缓冲液pH=6,酶用量为1mg/g,酶解温度50℃,酶法提取的提取率比传统提取法提高了37%。结论:该工艺简单、合理,适用于工业化生产。Objective To optimize enzymatic extraction process of Sophora alopecuroides L, in order to increase extracting rates of alkaloids. Methods With the content of alkaloids from Sophoraalopecuroides L as index, effects of enzyme dosage, enzymolysis time, pH and temperature on enzymatic extraction process were investigated by single factor test and orthogonal test. Results Its optimum extraction technology was as follows: enzyme dosage lmg/g, enzymolysis time of 4 hours, enzymatic hydrolysis of pH 6.0, enzymolysis temperature at 50℃ ; The extraction rate of enzymatic extraction technology was 1.34 times higher than the traditional technology. Conclusion This optimized extracting process is stable, reasonable and applicable to industrial production of Sophoraalopecuroides L.
