摘 要:目的:探讨MTT法和CCK-8法对细胞活性检测的最佳实验条件,并比较二者的优缺点。方法:取传至第5代的兔视网膜色素上皮细胞为研究对象,分别用MTT试剂和CCK-8试剂检测兔视网膜色素上皮细胞的增殖情况。结果:MTT法最佳检测波长为570nm,最佳检测时间在加入试剂后4h,细胞数量在1.25×103~2×104之间检测较好;CCK-8法最佳检测波长在450nm,最佳检测时间在加入试剂4~6h,适宜的细胞检测范围为1.25×103~1×104。结论:MTT法较CCK-8法细胞检测范围更广,孵育时间更短,是一种优于CCK-8法的细胞活性检测方法。Objective To investigate optimal experiment conditions between MTT assay and CCK -8 assay in rabbit retinal pigment epithelial cells, and compare the advantages and disadvantages of the two methods. Methods Rabbit retinal pigment epithelial cells of the fifth generation were studied. The activity of rabbit retinal pigment epithelial ceils were detected respectively with MTT reagent and CCK -8 reagent. Results For MTT assay, the best wavelength was 570nm, the best test was 4h after adding reagent, the number of rabbit retinal pigment epithelial cells within the range of 1.25 × 10^4 - 2 × 10^5 ; For CCK -8 assay, the best wavelength was 450nm, the best test was 4 - 6h after adding reagent, the suitable cell detection range is 1.25 × 10^4 - 1 × 10^5. Conclusion MTT assay is better than CCK - 8 assay in the aspects of detection range and incubation time, and MTT is superior to CCK - 8 in proliferation assay.
