摘  要:目的:研究AMPK蛋白分子在慢性间歇低氧幼鼠脑损伤中的作用机制。方法:取SPF级雄性健康SD幼鼠48只,随机分成4组:慢性间歇低氧2周组、4周组(2IH、4IH组),空气氧舱对照2周组、4周组(2AC、4AC)。用TUNEL(原位末端标记法)观察各组海马、皮层细胞凋亡情况;采用Western blot法检测各组海马、皮层AMPK磷酸化蛋白表达情况。结果:2IH组、4IH组海马和前额皮层区神经细胞凋亡指数明显高于空气对照组(各组均P<0.05)。2IH及4IH组海马和皮层AMPK磷酸化蛋白值均高于空气对照组(海马,皮层P<0.05)。结论:慢性间歇低氧可引起AMPK的磷酸化激活,并可诱导相关脑区神经元的凋亡,存在时间依赖性,提示p-AMPK可能在慢性间歇低氧幼鼠的脑损伤中起到关键作用。Objective:To investigate the role of AMPK-eEF2 signal pathway in brain injury following chronic intermittent hypoxia (CIH) in the weanling rat. Methods: Forty male Sprague-Dawley rats (3-week-old-4-week-old and 80 g to 100 g) were randomly divided into 4 groups: 2-week-CIH group (2IH), 4-week-CIH group (4IH), 2-week-simulated air control group (2C) and d-week- simulated air control group (4C). The morphologic changes were observed by HE staining and the cell apoptosis was detected with TUNEL staining. Then hippocampus and prefrontal cortexes were collected for expression analysis of AMPK by Western blot method respectively. Resluts :The neurons apoptosis in the hippocampus and prefrontal cortex in CIH exposed groups were more than control groups (P〈0.05, respectively), especially in 4IH group. The expression levels of AMPK protein in the hippocampus and prefrontal cortexes in IH groups were more than those in 2AC and 4AC groups (P〈O.05, respectively). Conclusion:Chronic intermittent hypoxia can up-regulate the AMPK transcription in brain regions which associated with learning and memory just like hippocampus and prefrontal cortex. In the end, the neuron apoptosis happened. Suggest that AMPK may play an important role in the learning-memory impairment of growing rats' exposure to chronic intermittent hypoxia.
