摘 要:目的分析干细胞因子(SCF)、单核细胞趋化蛋白1(MCP-1)在颅骨骨缺损模型大鼠中的作用及麝香对二者表达的影响。方法通过颅骨钻建立颅骨骨缺损模型,将模型大鼠随机分为给药组与模型组,再将给药组和模型组随机分为3小组,给药组灌服麝香,模型组灌服生理盐水,分别在干预后7 d、14 d、28 d通过酶联免疫吸附法(ELISA)检测各组大鼠血清中SCF表达及采用实时荧光定量PCR法检测颅骨骨缺损处MCP-1 mRNA含量的表达变化。结果与模型组比较,给药组第14天SCF表达最低,且无显著性,第7天和第28天其表达均比第14天升高,这两时间段与模型组比较差异均有统计学意义(P=0.0002、0.0025),但是第7天SCF表达增加更加显著;给药组在第7天MCP-1 mRNA表达水平最高,与模型组比较差异有统计学意义(P<0.05),第14天和第28天其表达持续降低,与模型组比较差异均无统计学意义(P=0.0633、0.1597)。结论SCF、MCP-1在颅骨骨缺损修复愈合过程中可能起重要作用,麝香通过增加模型大鼠血清中SCF的表达和颅骨骨缺损组织MCP-1 mRNA的表达,间接反映出骨缺损修复愈合机制可能与增加SCF和MCP-1 mRNA表达有关,麝香在加速骨缺损修复与愈合微环境中起着重要作用。Objective To analyze the effect of stem cell factor( SCF) and monocyte chemotaxis protein 1( MCP-1) on the skull bone defect and the effect of musk on the expression of the two factors in the rat model.Methods The skull bone defect model established with drilling.The model rats were randomly divided into treatment group and model group,and then the treatment group and model group were randomly divided into three groups.Rats in the treatment group were gavaged with musk,and rats in the model group were administrated with saline,respectively.After 7d,14 d,and 28 d of the intervention,the expression of SCF was detected with enzyme-linked immunosorbent sorbent assay( ELISA),and MCP-1 mRNA expression was detected with real-time fluorescence quantitative PCR.Results Compared with the model group,SCF expression was the lowest in the treatment group on the 14 th day,with no significant difference.SCF expression was higher at 7 days and 28 days than that at the 14 day,and the different was statistically significant between model group and treatment group( P = 0.0002,0.0025).The increase of SCF expression was more significantly on the 7th day.MCP-1 mRNA expression was the highest in the treatment group on the 7th day,and it was significantly different compared with the model group( P〈0.05).The expression continued to decrease on the 14 th and28th day,and it was not statistically significant compared with the model group( P = 0.0633,0.1597).Conclusion SCF and MCP-1 play an important role in bone defect healing.Musk increases the serum SCF expression and MCP-1 mRNA expression in the skull bone defect tissues,indirectly indicating that the healing of bone defects may be associated with the increase of SCF and MCP-1 mRNA expression.Musk plays an important role in accelerating the healing of bone defects in the microenvironment.
