摘  要:目的:利用硫酸软骨素酶(ChABC)将杏仁核的两个亚区基底外侧杏仁核(BLA)和中央杏仁核(CeA)内神经细胞周围基质网(PNNs)降解之后,观察对吗啡奖赏记忆形成过程的影响。方法:在大鼠BLA、CeA以及对照脑区BLA背侧2 mm分别微注射ChABC(0.2 U·ul^(-1))降解PNNs,给予大鼠2 d的吗啡(10 mg·kg^(-1),s.c.)诱导的条件性位置偏爱(CPP),通过CPP测试值来观察吗啡奖赏记忆的形成强度。结果:(1)与注射溶媒组相比,在BLA和CeA微注射ChABC组吗啡诱导的CPP值显著增高(P<0.05);(2)BLA背侧2 mm处微注射ChABC再进行CPP训练,吗啡诱导的CPP值在给药组和溶媒组之间没有显著性差异(P>0.05)。结论:本研究发现杏仁核PNNs的降解能够促进大鼠吗啡奖赏记忆的获得。Objective: To study the effect of degradation of perineuronal nets( PNNs) in basolateral amygdala(BLA) and central amygdala( CeA),two sub-regions of amygdala on the formation of morphine reward memory.Methods: We microinjected ChA BC( 0. 2 U·ul^-1) into the BLA,CeA or 2mm dorsal to the BLA in rats to degrade the PNNs; we trained rats with a consecutive 2-day conditioned place preference( CPP) procedure and used CPP value to study the strength of morphine reward memory.Results: The rats with microinjection of ChA BC into the BLA or CeA before morphine CPP training exhibited high CPP scores during the test,compared with vehicle group( P〈0. 05). However,the rats with microinjection of ChABC into the control region outside the amygdala,2 mm dorsal to the BLA had a similar morphine CPP value compared with the vehicle group( P〉0. 05).Conclusion: Results indicate that PNNs degradation in amygdala facilitated new learning of morphine CPP,supporting the evidence that PNNs / CSPGs degradation in amygdala is critical for acquisition enhancement of drug reward memory.
