摘 要:目的探讨SLC34A2对肺癌肿瘤干细胞(carcinoma-initiating cells,CICs)侵袭和增殖作用的影响及机制。方法采用流式细胞仪分选技术从肺癌细胞株H1650中分选出CD44+/CD133+的CICs;LV3-SLC34A2组沉默SLC34A2基因,LV3-NC组为对照。使用绿色荧光蛋白及免疫印迹法检测SLC34A2沉默效率及沉默效果;沉默SLC34A2后,免疫印迹法检测SLC34A2编码蛋白Na Pi-Ⅱb的表达,使用Transwell侵袭实验检测对CICs侵袭能力的影响,肿瘤细胞成球实验检测对CICs增殖能力的影响,免疫印迹法检测JAK/STAT信号通路相关蛋白的表达情况。结果与LV3-NC组比较,LV3-SLC34A2组中Na Pi-Ⅱb蛋白的表达水平下降(P<0.05)。沉默SLC34A2可以有效抑制肺癌CICs的侵袭和增殖能力(P<0.05),并使JAK/STAT信号通路中的P-JAK和P-STAT蛋白表达相应降低(P<0.05)。结论沉默SLC34A2基因可能通过调控JAK/STAT信号通路来抑制肺癌CICs的侵袭和增殖作用。Objective To investigate effects of SLC34A2 on invasion and proliferation of human lung carcinomainitiating cells (CICs) and its mechanism. Methods CD44 +/CD133 + CICs in lung cancer were selected from lung cancer cell line H1650 by flow cytometry sorting technique. In LV3-SLC34A2 group, SLC34A2 gene was silenced, while in LV3-NC group, LV3-NC was used as control. Green fluorescent protein (GFP) and western blotting methods were used to detect the efficiency and effectiveness of SLC34A2gene silencing. After silencing SLC34A2, western bohting method was used to detect expression of SLC34A2 coding protein NaPi-Ⅱb, and its invasion ability on CICs was detected by Transwell matrigel invasion assays. Effect on CICs proliferation ability was detected by tumor cells in ball assays. Related protein expressions of JAK/STAT signal pathway were detected by Western blotting method. Results Compared with that in LV3-NC group, expression of NaPi- IIB protein was significantly decreased in LV3-SLC34A2 group (P 〈 0.05 ). Silencing SLC34A2 effectively inhibited the invasion and proliferation of CICs (P 〈 0.05 ) , and it could decrease the protein expression levels of P-JAK and P-STAT proteins in JAK/STAT signal pathway (P 〈 O. 05 ). Conclusion Si- lencing SLC34A2 can inhibit the effects of CICs invasion and proliferation by regulating JAK/STAT signal pathway.
