摘 要:目的探讨EZH2基因与慢性髓细胞白血病(CML)的关系。方法收集慢性髓细胞白血病(CML)40例;以非恶性血液病患者20例(ctrl组)作为对照。收集患者外周血标本,分离单个核细胞,提取RNA,采用实时荧光定量PCR(Real-Time PCR)分别检测慢性髓细胞白血病(CML)组和对照组EZH2基因表达情况。同样分离单个核细胞,分别裂解,提取蛋白,利用蛋白质印迹法(Western Blot)检测EZH2蛋白在各组中的表达,并与疾病的临床特征进行相关性分析。结果与对照组相比,白血病组的EZH2基因表达水平明显上调(P<0.05),且EZH2蛋白水平也明显升高。结论 EZH2基因与慢性髓细胞白血病(CML)的发生发展有密切关系。Objective To investigate the relationship between EZH2 gene and chronic myelogenous leukemia. Methods Sixty newly - diagnosed patients were enrolled in this study, and divided into two groups : including chronic myelogenous leukemia (CML) 40 cases, and 20 patients without malignant hematological diseases were the control (ctrl group). We collected the peripheral blood samples of patients, isolated the mononnclear cells, purified the RNA and used real - time fluorescent quantitative PCR ( Real - Time PCR) to detect the gene expression of EZH2 in chronic myelogenous leuke- mia (CML) group and control group. Equally, we isolated the mononuclear cells, purified the protein and used Western Blot to detect the protein expression level of EZH2 in each group. Then analyzed their correlation with the chronic my- elogenous leukemia patients. Results Compared with control group, the expression levels of PTEN gene in chronic my- elogenous leukemia (CML) group were significantly increased (P 〈 0. 05 ) while the protein expression levels of EZH2 were significantly increased too. Conclusion The EZH2 genes were closely related to the prognosis of chronic myeloge- nous leukemia (CML).
