摘  要:目的:体外观察白细胞介素17(interleukin17,IL-17)对小鼠髓源抑制性细胞(myeloid-derived suppressor cells,MDSCs)凋亡的影响。方法:通过皮下注射Lewis细胞构建荷瘤小鼠模型,采用免疫磁珠分选技术(MACS)分离脾脏MDSCs,常规培养。经IL-17处理24 h后,采用流式细胞术检测MDSCs凋亡率;经IL-17处理6 h后通过蛋白质印迹法检测细胞Bcl-2的表达。结果:IL-17处理组活细胞数量为(9.547±0.962 6)×10~4,明显高于对照组活细胞数[(4.123±0.259 8)×10~4,P<0.01];IL-17处理组活细胞比例为(59.55±2.831)%,明显高于对照组[(45.33±1.556)%,P<0.05];对照组晚期凋亡细胞比例为(22.87±1.349)%,明显高于IL-17处理组[(13.70±1.003)%,P<0.01]。蛋白质印迹检测结果显示,IL-17处理后的MDSCs抗凋亡蛋白Bcl-2表达水平明显上调(P<0.05)。结论:IL-17体外可明显抑制MDSCs的凋亡。Objective: To observe the apoptosis of myeloid-derived suppressor cells (MDSCs) that is treated by IL-17 and explore the effect of IL-17 on apoptosis of MDSCs. Methods: Lewis lung carcinoma (LLC) tumor bearing models were constructed with subcutaneous injection of Lewis cells. MDSCs were isolated from spleen in tumor-bearing mice and cultured in RPMI 1640 culture media. After treating MDSCs with IL-17 for 24 h, apoptosis of MDSCs were detected by Annexin V and PI. Furthermore,Western blotting was also used to detect the expression of anti-apoptotic protein Bcl-2 after treating MDSCs with IL-17 for 6 h. Results: The counts of viable cells of MDSCs were(4. 123 ± 0. 259 8) × 104 in control group and (9. 547 +0. 962 6) × 104 in IL-17 treatment group (P 〈0.01 ) ; percentage of viable cells of MDSCs was (45.33 ±1.556)% in control group and (59.55 ±2. 831)% in IL-17 treatment group (P 〈0.05). The later apoptosis of MDSCs were (22.87 ±1. 349) % in control group and ( 13.70 ± 1. 003 ) % in IL-17 treatment group(P 〈0.01 ). Results of Western blotting showed that the expression level of anti-apoptotic protein Bcl-2 significantly increased after IL-17 treatment(P 〈 0.05 ). Conclusion: IL-17 may inhibit the apoptosis of MDSCs in vitro significantly.
