摘  要:目的检测Fra-1在肝细胞性肝癌(简称肝癌)及癌旁组织中的表达情况,探讨其与肝癌病理特征间的关系。方法采用免疫组化法检测Fra-1在60例肝癌及癌旁组织中的表达情况;采用Western blot、实时定量PCR(qRT-PCR)法检测Fra-1在20对冰冻肝癌及癌旁组织中的相对表达量。结果免疫组化示Fra-1在肝癌组织细胞核中阳性表达率为57%(34/60),细胞质中未见明显表达;而Fra-1在癌旁组织细胞质阳性表达率为23%(14/60),细胞核中未见明显表达。Fra-1在肝癌组织中的表达和血清甲胎蛋白(sAFP)、肿瘤血管侵犯相关(χ~2=4.538、9.086,P<0.05),而与肝癌患者的肿瘤包膜、肝癌TNM分期、Edmondson分级等其他临床病理特征无明显相关性;Western blot及qRT-PCR检测结果示肝癌组织中Fra-1的相对表达量高于癌旁组织(P<0.01)。结论 Fra-1与sAFP及肿瘤血管侵犯相关,提示Fra-1可能参与调控肝癌甲胎蛋白(AFP)的生成并影响肝癌的发展;为肝癌的诊治提供一些新的思路及理论基础。Objective To detect the expression of Fra-1 in hepatocellular carcinoma(HCC) tissues and their corresponding peritumorous tissues,as well as its clinical significance. Methods Immunohistochemical staining was used to detect the expression of Fra-1 in 60 cases of HCC tissues and their corresponding peritumorous tissues.Western blot and real-time quantitative PCR(qRT-PCR) were used to assess the relative expression levels of Fra-1in 20 pairs of frozen HCC tissues and their corresponding peritumorous tissues. Results Immunohistochemical staining showed that the positive expression rate of Fra-1 in the nucleus of the 60 HCC tissues was 57%,which appeared almost 0 in the cytoplasm. The positive expression rate of Fra-1 in the cytoplasm of the peritumorous tissues was 23%,which appeared almost 0 in the nucleus. The expression of Fra-1 in tumor cells was related to sAFP(serum alpha fetal protein) and vascular invasion(χ~2= 4. 538,9. 086,P〈0. 05),while not to the other clinicalpathological features,such as tumor capsule,TNM stage and Edmondson classification. The results of Western blot and qRT-PCR showed that the expression level of Fra-1 in HCC tissues was higher than their corresponding adjacent tissues(P〈0. 01). Conclusion Fra-1 is related to sAFP and vascular invasion,which shows that Fra-1 may be involved in the formation of AFP and affect the development of HCC. It may provide some new insight and theoretical basis for the dignosis and treatment of HCC.
