摘　　要：目的:探讨微小核糖核酸(miRNA)-506对人直肠癌Colo320细胞增殖和侵袭的影响。方法:构建miRNA-506模拟物,应用脂质体法转染直肠癌细胞株Colo320。采用逆转录酶链聚合反应(qRT-PCR)法检测各组细胞中miRNA-506的表达水平,CCK-8法检测细胞的增殖情况。Transwell小室检测细胞的侵袭能力。Western blot检测MMP-9的表达。结果:qRT-PCR结果显示,miRNA-506转染组miRNA-506的表达量明显高于空白细胞对照组和阴性对照组(P均<0.05)。CCK-8结果显示,上调miRNA-506抑制Colo320细胞增殖。Transwell结果显示,转染miRNA-506后Colo320细胞的侵袭数目明显减少,差异具有统计学意义(P<0.05)。miRNA-506可以抑制Colo320细胞MMP-9蛋白的表达。结论:上调miRNA-506能抑制人直肠癌Colo320细胞的增殖和侵袭能力。Objective：To investigate the effect of microRNA-506 on the proliferation and invasion of human rectal cancer Colo320 cells.Methods：Synthetic miRNA-506 and its negative control were transfected into Colo320 cells by liposome method.After transfection,the expression of miRNA-506 level was detected by quantitative real-time PCR.The proliferation of Colo320 cells was detected by CCK-8 assay,and the invasion ability changes were analyzed in transwell.The expression of MMP-9 was detected by Western blot.Results：The results of RT-PCR showed that the expression of miRNA-506 in transfection group were up-regulation than control group.The proliferation of Colo320 cells were significantly inhibited by up-regulation miRNA-506.The invasion of Colo320 cells were significantly inhibited by up-regulation miRNA-506.Conclusion：miRNA-506 could suppress the proliferation and invasion of rectal cancer Colo320 cells.
