多重PCR试剂盒在检测致泻性大肠埃希菌中的应用

摘　　要：目的应用商品化多重PCR试剂盒来检测致泻性大肠埃希菌,以建立更快速、高效的致泻性大肠埃希菌监测工作方法。方法应用某市场商品化致泻性大肠埃希菌多重PCR试剂盒来初筛检测福建省的腹泻病分离的菌株,用本实验室前期已建立的致泻性大肠埃希菌不同的PCR方法进行串联试验对初筛结果加以验证。结果验证试验从2015年福建省2个监测点分离的经生化反应初步鉴定为大肠埃希菌的菌株中,致泻性大肠埃希菌鉴定率为7.00%(21/300),阳性菌株为21株,均成功获得了分纯单克隆菌株。结论商品化致泻性大肠埃希菌多重PCR试剂盒对于提高效率、减少繁重的工作量有帮助。Objective To develop a more rapid and efficient method for the detection of diarrheagenic Escherichia coli,a commercial multiplex PCR kit was applied and evaluated. Methods A commercial diarrheagenic Escherichia coli multiplex PCR kit was applied in the preliminary screening for the strains isolated from diarrheal diseases patients in Fujian province. Monoclonal colonies were selected after the suspicious positive strains were inoculated on plates,then,different PCR methods of diarrheagenic Escherichia coli early established by our laboratory was applied as serial testing for comfirmed suspicious positive results.Results Among the Escherichia coli strains identified with test from 2 monitoring sites in Fujian province in 2015,7. 00% of diarrheagenic Escherichia coli were identified（ 21/300）,and pure monoclone was successfully obtained of the 21 strains. Conclusion The commercial multiplex PCR kit of diarrheagenic Escherichia coli was helpful to improve the efficiency and reduce the heavy workload.
