摘  要:目的:探讨白藜芦醇对人白血病细胞增殖的影响及其机制研究。方法:采用不同浓度白藜芦醇(resveratrol,Res)及伊马替尼(imatinib,IM)对K562细胞进行干预,流式细胞术观察细胞凋亡情况,采用荧光定量PCR及Western blot检测其给药后COX-2、b FGF、TGFβ1及VEGF的变化。结果:流式细胞术检测显示单用白藜芦醇及单用伊马替尼对人白血病细胞K562均具有凋亡作用,而随着白藜芦醇浓度的增加,K562细胞的凋亡率逐渐增高;随着伊马替尼浓度的增加,K562细胞的凋亡率逐渐上升。PCR及Western blot结果显示,与对照组相比,0.2μmol/L伊马替尼组的COX-2、b FGF及VEGF的蛋白表达量及mRNA表达水平明显降低,而TGFβ1的蛋白表达量及mRNA表达水平显著增高,差异具有统计学意义(P<0.05);而用200μmol/L白藜芦醇+0.2μmol/L伊马替尼联合作用于K562细胞后,与不用药对照组和0.2μmol/L伊马替尼组比较,细胞中TGFβ1的蛋白表达量及mRNA表达水平显著增高,而COX-2、bFGF及VEGF的蛋白表达量及mRNA表达水平均显著降低(P<0.05)。结论:白藜芦醇及伊马替尼对白血病K562细胞株均具有促进凋亡作用,联合用药可明显增加其凋亡率。白藜芦醇抗白血病K562细胞作用机制可能是通过下调COX-2、bFGF及VEGF mRNA和蛋白的表达及上调TGFβ1 mRNA和蛋白的表达来实现的。Objective: To investigate the mechanism influencing the growth of leukemic cells. Methods: Different concentrations of resveratrol and imatinib treated K562 cells were examined by flow cytometry apoptosis. Fluorescence quantitative PCR and Western blot detected the changes of COX - 2, bFGF,TGFpl and VEGF after its administra-tion. Results: Flow cytometry showed that resveratrol alone and single with imatinib on human leukemia cell K562 had apoptosis, and with the increase of the concentration of resveratrol, K562 cells apoptosis rate gradually increased, with the increase of imatinib concentrations, K562 cells apoptosis rate gradually increased. PCR and Western blot analysis showed that, compared with the control group , the protein and mRNA level of COX - 2 , bFGF and VEGF, in the 0. 2μmol/L imatinib group,decreased significantly,while the level of TGFpl protein and mRNA expression signifi-cantly increased. The difference was statistically significant ( P 〈0. 05 ) . However, with 200μmol/ml resveratrol + 0. 2μmol/L imatinib acting on the K562 cells,the level of TGFβ1 protein and mRNA expression was significantly increased,while the expression of COX-2 ,bFGF and VEGF was significantly reduced,and a statistically significantdifference was observed when compared with control group and the 0. 2μmol/L imatinib group (P 〈0. 05) . Conclu-sion : Resveratrol and imatinib on leukemia cell line K652 could promote apoptosis,which may be associated with a significantly increased rate of apoptosis. Resveratrol acted anti - leukemia has effects on K562 cells, which may be through decreasing the expression of COX -2,bFGF,VEGF mRNA and protein, increasing the expression of TGFβ1 mRNA and protein.
