摘  要:目的探讨HGF对紫外线诱导成纤维细胞产生ROS的影响。方法采用cck-8法检查不同浓度HGF对成纤维细胞(FBs)活性的影响。HGF预处理FBs 2h,装载ROS检测探针,UVB照射诱导体外成纤维细胞发生氧化应激,荧光显微镜、酶标仪分别检测ROS的表达。结果 5ng/ml HGF对成纤维细胞的增值无明显影响。在UVB诱导的应激性老化中,ROS表达显著升高(<0.01)。5ng/ml HGF能显著降低UVB引起的ROS表达(<0.05)。结论 HGF可降低UVB造成的光老化FBs中ROS的表达,减少氧化应激反应,可作为对抗光老化的新手段。Objective To explore the effect of hepatocyte growth factor (HGF) on reactive oxygen species (ROS)in UVA-induced skin fibroblasts. Methods The effect of HGF of different dosage on the activity of fibroblasts (FBs) wasdetected by cck-8 assay; after HGF pretreatment for 2 hours, ROS test probe was loaded and oxidative stress of FBs wasinduced by UVB irradiation in vitro; the expression of ROS was measured by fluorescence and enzyme-labeling instrumentrespectively. Results 5ng/ml HGF had no obvious effect on FBs proliferation; the expression of ROS increased greatly inUVB-induced oxidative stress( P〈0.01); 5ng/ml HGF reduced the ROS expression caused by UVB induction( P〈0.05).Conclusions HGF can reduce ROS expression in FBs induced by UVB irradiation and decrease oxidative stress, it can be takenasa new anti-photoaging measure.
