摘  要:目的:研究干扰素诱导蛋白AIM2(absent in melanoma 2)在乳腺癌细胞系及乳腺癌组织中的表达及意义。方法:建立MCF-7 t TA-AIM2细胞为实验组,MCF-7 t TA-Luc细胞为对照组。以RT-PCR分析在不同的乳腺癌细胞株以及乳腺癌组织中AIM2基因的表达。利用TUNEL检测细胞凋亡。结果:本实验建立的Tet-off诱导系统可使t TA结合到TRE,从而促进AIM2基因转录,进而使AIM2蛋白能大量表达。AIM2基因表达量在MCF-10A、184A1细胞株中非常低,在MDA-MB-435中最高。同时,乳腺癌组织较邻近正常乳房组织有较多AIM2基因表达。AIM2可促进乳腺癌细胞凋亡。结论:在乳腺癌Tet-off诱导系统中,AIM2可促进乳腺癌细胞凋亡,同时在MDA-MB-435及乳腺癌组织中的表达较高。Objective: To study the expression and significance of absent in melanoma AIM2 in breast cancer cell line and breast cancer tissue. Methods: MCF-7 tTA-AIM2 cells and MCF-7 tTA-Luc were established as the experimental group and control group respectively. RT-PCR was used to analyze the AIM2 gene expression in different breast cancer cell lines and in breast cancer tissue. Apoptosis was detected by staining with TUNEL. Results: This experiment established a Tet- off guidance system which can make the tTA to TRE, promote AIM2 gene transcription, and upregulate the AIM2 protein multiplicity expression. AIM2 gene expression data in MCF-10A, 184A1 cell lines was very low, and the highest in the MDA-MB-435. At the same time, the breast tissues have more AIM2 gene expression than the adjacent normal breast tis- sues. AIM2 expression can significantly promote the apoptosis of breast cancer ceils. Conclusion: In breast cancer in- duced by Tet-off system, AIM2 can promote breast cancer cells apoptosis, at the same time in MDA-MB-435 and in breast cancer tissue has high expression.
