摘  要:目的:使用响应面优化法,确定从西洋参茎叶中提取多糖的最佳提取工艺,并且探讨西洋参茎叶多糖的抗病毒活性。方法:设计单因素实验,并在单因素实验的基础上进行响应面实验设计,确定最佳提取工艺。提取的多糖样品,以利巴韦林为对照,RSV-Hep2细胞体外病毒感染模型进行体外抗RSV作用的对比实验。结果:经优化实验得到的最佳工艺为:提取时间为90 min,提取次数为2次,料液比为1:35,最佳优化结果所得多糖的抗病毒TI指数为32.57。结论:在使用响应面软件优化下,确定了最佳提取工艺,并且发现得到的茎叶多糖具有抗病毒作用,对西洋参茎叶多糖的进一步研究提供依据。Objective:To extract polysaccharide from Panax quinquefolius leaves and stems,determine the preparation technology of polysaccharide by Response Surface Methodology and discussed the antiviral activity of polysaccharide from Panax quinquefolius stems and leaves. Method:Sample was refluxed with water in the water bath and determination of polysaccharide extraction rate and content. Design single factor experiment,on the basis of preliminary experimen,response surface design was applied by design expert to find the optimum extracting conditions. Then using the ribavirin for reference substance and utilizing the RSV-Hep2 cells as in vitro infection model to compare the anti RSV effects in vitro. Results:After analysis of the preliminary experiment,the best extracting conditions was as follows:extraction time is 90 min,extraction frequency is 3 times,the ratio between drug and liquid is 1∶35,the The highest extraction yield of polysaccharide was 1.86%,The TI index of the polysaccharide were 32.57. Conclusion:With the optimization of response surface design,determined the preparation technology of polysaccharide and found that the plant polysaccharides have antiviral effect. Provide the basis for further study of polysaccharide from Panax quinquefolius leaves and stems.
