摘  要:目的:研究自发性2型糖尿病(T2DM)大鼠ZDF(Zucker diabetic fatty)(fa/fa)与ZDF(fa/+)及经糖耐康干预后,ZDF(fa/fa)肠道共生细菌菌群结构的变化。方法:将6只雄性ZDF(fa/+)大鼠设为正常组,雄性ZDF(fa/fa)大鼠18只(不同日随机血糖均值≥11.1 mmol·L^(-1))设为T2DM成模组。根据体重及随机血糖(RBG)随机分为3组,分别为模型组(生理盐水10 mL·kg^(-1)),糖耐康高、低剂量(糖耐康3.24,0.81 g·kg^(-1))组,每组6只。给药6周后取材,腹主动脉取血检测大鼠空腹血糖(FPG)及空腹血清胰岛素(FINS);收集大鼠肠道内粪便,行逆转录-聚合酶链式反应(RT-PCR),通过扩增子焦磷酸测序分析16S rRNA V4区菌群的改变。结果:与模型组比较,糖耐康高、低剂量组大鼠FPG,FINS有显著改善(P<0.01)。各组大鼠在菌群丰度多样性及结构组成上有显著差异。在科及以下水平上,与正常组比较,模型组乳酸杆菌明显升高(P<0.05);与模型组比较,糖耐康高、低剂量组乳酸杆菌丰度有所下降,但无统计学差异,普雷沃氏菌丰度显著升高(P<0.01),且普雷沃氏菌属与ZDF大鼠FINS呈正相关。结论:ZDF(fa/fa)与ZDF(fa/+)大鼠在肠道菌群结构上有很大差异,糖耐康对ZDF大鼠血糖的改善作用可能是通过影响ZDF大鼠肠道中乳酸杆菌、普雷沃氏菌的丰度实现。Objective: To investigate the changes in gut microbiota structure after Tangnaikang (TNK)intervention in Zucker diabetic fatty rats (ZDF, fa/fa) and ZDF (fa/+ ) with spontaneous type 2 diabetes mellitus. Method: Six male ZDF (fa/+ ) rats were set as normal control group, and eighteen male (fa/fa) ZDF rats (mean value of random blood glucose in different days ≥ 11.1 mmol·L-1) were set as T2DM model group and divided into model group (normal saline, 10 mL·kg-1) , TNK high-dose (TH, 3.24 g·kg-1) and TNK low-dose (TL, 0. 81 g·kg-1) groups depending on their body weight and random blood glucose (RBG), n = 6 in each group. After 6 weeks of drug administration, blood was taken from abdominal aorta to detect their fasting plasma glucose (FPG) and fasting serum insulin (FINS), fecal samples were collected from the rats to detect microbiota by reverse transcription-polymerase chain reaction (RT-PCR) , and amplicon pyrose quencing was taken to analyze the changes of microbiota in 16S rRNA V4 region. Result: Both TH group and TL group had significant differences in FPG and FINS as compared with model group (P 〈 0.01 ). There were significant differences in flora diversity and structure composition between various groups. At the family level and below, the abundance of Lactobacillaceae was increased in model group as compared with control group (P 〈 0.05) ; while as compared with model group, the abundance of Lactobacillaceae was decreased in TH group and TL group without statistical difference, and the abundance of Prevotella was significantly increased (P 〈 0.01 ) , in addition, Prevotella had a positive correlation with FINS in ZDF rats. Conclusion: The ZDF (fa/fa) and their lean llittermates ZDF (fa/+ ) had significant differences in microbial composition, and TNK may improve the blood glucose by affecting the abundance of Prevotella and the Lactobacillaceae in intestine of the ZDF rats.
