摘  要:目的探讨芥酸酰胺对小鼠的抗焦虑样作用及其机制。方法小鼠每天1次芥酸酰胺灌胃给药,连续7 d。第7天给药后1 h进行高架零迷宫实验或先施行小鼠强迫游泳应激后立即进行水迷宫实验,行为学实验结束后断头迅速取双侧海马备用。分别采用比色法和酶联免疫吸附法(enzyme linked immunosorbent assay,ELISA)测定海马组织单胺氧化酶(monoamine oxidase,MAO)活性、去甲肾上腺素(norepinephrine,NE)和5-羟色胺(5-hydroxytryptamine,5-HT)含量。结果芥酸酰胺显著增加小鼠高架零迷宫和水迷宫实验中的开臂进入次数、开臂滞留时间及缩短寻找目标的潜伏期;降低海马组织中MAO活性,增高NE和5-HT的含量。结论芥酸酰胺对小鼠可能有一定的抗焦虑样作用,其机制可能与调节海马组织单胺类神经递质有关。Objective To investigate the anxiolytic like effects of erucamide on mice and its possible mechanism.Methods Erucamide(5,10,20 mg·kg-1) was administered to mice by gavage for 7 consecutive days once daily.Mice were induced stress by forced swimming treatment one hour after the last administration,and then the mice were immediately submitted the elevated zero maze and water maze test.After behavior tests,the bilateral hippocampal tissues were quickly removed for subsequent biochemical indicator detection.The activities of monoamine oxidase(MAO),contents of norepinephrine(NE) and 5-hydroxytryptamine(5-HT) in hippocampus were determined by the colorimetric method and enzyme linked immunosorbent assay(ELISA).Results As compared with the model group,the open arm entries and the open arms retention time were significantly increased in the elevated zero maze,the latency for finding the target was reduced in the water maze test;and MAO activity was decreased,NE and 5-HT contents were increased in the hippocampus of erucamide treatment group.Conclusions Erucamide may have some anxiolytic like effects in mice;its mechanism may be related to inhibition of monoamine oxidase activity and regulation on monoamine neurotransmitters in hippocampus.
