莪蚕健胃方调控MMP-2表达对胃癌前病变血管生成作用的研究

摘　　要：[目的]探讨莪蚕健胃方对胃癌前病变大鼠MMP-2蛋白及其mRNA对血管生成的影响。[方法]实验大鼠分为正常组5只,造模组55只。制作胃癌前病变模型24周并抽检5只确保成功后,将剩余模型大鼠随机分为模型组,维甲酸组和中药高、中、低剂量组,每组10只。给药16周后处死取材。进行胃组织病理检测,MVD计数,和MMP-2蛋白及mRNA的检测。[结果]与正常组比较,造模组的MVD和MMP-2蛋白及其mRNA均过度表达(P<0.05),并随异型增生级别的升高而增强。MMP-2蛋白与MVD正相关(r=0.560,P<0.05),MMP-2mRNA与MVD无相关性(r=0.297,P>0.05)。在MVD、MMP-2蛋白及其mRNA的表达方面:与模型组相比,大部分治疗组能抑制其过度表达(P<0.05),中药低剂量组(P>0.05)除外。与维甲酸组相比,中药高剂量组表达降低(P<0.05),中药中剂量组差异无统计学意义(P>0.05)。[结论]1血管生成在胃癌前病变阶段即出现,MMP-2蛋白、mRNA呈过度表达,均随异型增生程度加重而增强。2大鼠胃癌前病变组织中MMP-2仅在蛋白水平与MVD呈正相关,提示MMP-2主要在蛋白层面参与了血管生成。3莪蚕健胃方可抑制胃癌前病变的血管生成,可能是通过削弱MMP-2基因的转录及表达而发挥治疗作用的。其作用与剂量有关。[Objective]To investigate the effects on angiogenesis from Ecan Jianwei recipe of gastric pre- cancerous lesions rats＇MMP-2＇s protein and mRNA. [Methods]The experimental rats were divided into normal group with 5 rats,the module group with 55 rats. And sampled 5 rats to ensure the success of making gastric precancerous lesion model for 24 weeks, then divided the rest of the model rats randomly into the model group, the retinoic acid treatment group, the high dose of Chinese medicine, medium dose. low dose treatment groups with 10 each. Administering for 16 weeks then obtain specimens. Carried out pathological detection of stomach tissue and evaluated the MVD and the level of MMP-2＇s protein and mRNA. [Results]Comparing with normal group,MVD and MMP-2＇s protein.mRNA of model groups were exces- sive expressed（P〈0.05）and them increased with the hyperplasia＇ s degree. There was a positive correlation between MVD and MMP-2＇s protein（r = 0. 560, P（0.05）, but showed no correlation between MMP-2＇s mRNA and MVD（r=0. 297, P〉0.05）. Interms of MVD.the expression of MMP-2＇s protein and mRNA： Comparing with model group,most treatment groups can restrain their excessive expressions（P〈0.05）ex-cept the Chinese medicine low dose group（P〉0. 05）. Comparing with retinoic acid group, the Chinese med- icine high dose group＇s expression was lower（P〈0.05）, the Chinese medicine middle dose group was no statistically significant difference（P〉0. 05）. [Conclusion] （1） Angiogenesis had started in gastric precancer- ous lesions, MMP-2＇s protein and mRNA showed overexpressions, all of them increased with the hyperpla- sia＇s degree. （2）In gastric precancerous lesions rats, MMP-2 were positively correlated with MVD at the protein level only,which indicated that MMP-2 participated in the angiogenesis mainly in protein level. （3） Eean Jianwei recipe played a therapeutic role by undermining MMP-2＇s gene transcription and expression to inhibit angiogenesis in gastric prec
