摘 要:为了研究中药辣蓼中总黄酮的提取工艺及其抗菌活性。采用均匀设计试验法,以总黄酮含量为响应值,考察了乙醇浓度、料液比、提取时间、提取温度对其影响,用DPS数据处理系统软件进行结构分析;采用微管-平板法测定辣蓼提取物对金黄色葡萄球菌的抗菌活性。结果显示,辣蓼总黄酮的提取工艺条件为:料液比1:60,乙醇浓度70%,提取时间1 h,提取温度为50℃,在该条件下,辣蓼中总黄酮含量为5.35%±0.23%,辣蓼总黄酮对金黄色葡萄球菌的最小抑菌浓度为0.782 mg/mL。该提取工艺操作简便,可用于中药辣蓼中总黄酮的提取且提取物对金黄色葡萄球菌有抗菌活性。In order to study the extraction process of total flavonoids from Polygonum hydropiper in traditional Chinese medicine and its antibacterial activity, 4 factors included the ethanol concentration, solid-liquid ratio, extraction time, extraction temperature, were studied by the uniform design method and the content of total flavonoids were as the index. The extraction process parameters were analyzed by 9.5 version DPS software. Theantibacterial activityto staphylo-coccus aureus was detected by the microtubule plate method. The results showed that the extraction process of total flavonoids was ethanol concentration of 70%, extract time of one hour, extract temperature of 50 ℃ and solid-liquid ratio of 1:60. Under these conditions, the content of total flavonoids from Polygonum hydropiperwas 5.35%±0.23%. The minimum inhibitory concentration of total flavonoids against Staphylococcus aureus was 0.782 mg/mL. So this extraction process was simple and could be used for extraction of total flavonoids from Chinese medicine polygonum hydropiper. And the extraction had antibacterial activity against staphylococcus aureus.
