摘 要:为明确抱茎苦荬菜内生细菌G-5分类地位及其抗菌蛋白在生物农药开发中的应用潜力,本研究采用16S rRNA、硫酸铵沉淀法、生长速率法和琼脂扩散法对其分类地位和抗菌蛋白抑菌特性进行分析。结果表明,内生细菌G-5鉴定为Bacillus methylotrophicus;抗菌蛋白对番茄早疫病病菌、梨黑斑病病菌和番茄灰霉病病菌具有较好抑菌效果,抑菌率分别为88.3%、90.5%和95.4%;最佳硫酸铵沉淀饱和度为80%;在温度小于80℃、pH 5~10和紫外照射12h后仍有较好抑菌活性。该抗菌蛋白在生物农药开发中具有一定的应用潜力。In order to understand application potential of endophytic bacteria G-5 from lxeris sonchifolia Hance in the development of bio-pesticide, taxonomic status and antifungal characteristic were studied by 16S rRNA, grade- precipitating with ammonium sulfate, plate growth rate method and agar diffusion method. The results showed that strain G-5 was identified as Bacillus methylotrophicus. Antifungal protein showed significantly antifungal activity against Alternaria solani, Alternacia alternate and Botrytis cinerea, with inhibition rate of 88.3%, 90.5% and 95.4%, respectively. Besides, optimal ammonium sulfate precipitation saturation was 80%, and the protein also had much better antifungal activity at the temperature bellow 80 ℃, pH 5-10 and UV irradiation for 12 hours. So, it indicated that the antifungal protein had certain application potential in bio-pesticide development.
