摘 要:采用中心组合设计实验和响应面分析法,开展红球菌SY095产生物表面活性剂70L罐的发酵工艺优化,并对提取的生物表面活性剂性质进行研究。通过研究发酵主要影响因子pH和溶氧交互作用,获得生物表面活性剂的最佳发酵方案为:pH 7.89,溶氧32.56%。在该条件下,发酵30h生物表面活性剂粗品产量达17.55g/L,发酵液表面张力从初始的59.235m N/m下降到26.455m N/m。红球菌SY095产生物表面活性剂对环境具有较强适应性,在高温(50-100℃)、高盐(NaCl浓度12%)和较宽pH(2-10)条件下都能保持较高的表面活性,稳定性良好。The large-scale fermentation process in 70 liter fermenter culture of biosurfactant producing by Rhodococcus SY095 was optimized by central composite design and response surface methodology, and the properties of the extracted biosurfactant were studied. By studying the pH and dissolved oxygen interactions which were the main influence factors, the optimum larger-scale fermentation conditions for the biosurfactant were determined as follows: pH 7.89, dissolved oxygen 32.56%. Under the optimum condition, the yield of biosurfaetant could reach 17.55g/L for 30 hours and the surface tension of fermentation supernatant could reduce from culture broth 59.235mN/m to 26.455mN/ m. The biosurfactant producing by Rhodococcus SY095 has strong adaptability to environments. It also demonstrated high surface activity under temperature(50-100℃), sah(12% NaC1 concentration) and wide adaptable pH(range of 2-10).
