奥氮平对MK-801处理大鼠自发活动和额叶微小RNA-219表达的影响

摘  要:目的:探讨非典型抗精神病药奥氮平对N-甲基-D-天门冬氨酸(NMDA)受体拮抗剂MK-801处理大鼠自发活动和额叶微小RNA(miRNA)-219表达的影响。方法:将18只成年雄性SD大鼠随机分为对照组(生理盐水(NS)灌胃+NS腹腔注射)、MK-801组(NS灌胃+MK-8010 1 mg/kg腹腔注射)及奥氮平组(奥氮平2mg/kg灌胃+MK-8010 1 mg/kg腹腔注射):每日给药1次,1周后做开场实验检测大鼠自发活动,用实时荧光定量PCR技术检测各组大鼠额叶miRNA-219的表达。结果:与对照组比,MK-801组大鼠自发活动增加,额叶miRNA-219表达下降,差异均有统计学意义(P<0.01);而与MK-801组比,奥氮平组大鼠自发活动减少,额叶miRNA-219表达增加,差异有统计学意义(P<0.05或P<0.01)。结论:奥氮平能抑制大鼠自发活动的增加及额叶miRNA-219的下调。Objective:To investigate the effects of olanzapine on locomotor activity and the expression of microRNA (miR) -219 in frontal cortex of rats treated by a N-methyl-D-aspartate(NMDA) receptor antagonist, MK-801. Methods: 18 adult male SD rats were randomly divided into three groups(n=6):control group(salinei.g.+salinei.p.), model group(saline i.g.+MK-801 0. lmg/kg i.p.),olanzapine+MK-801 group(olanzapine 2mg/kg i.g.+MK-8010. lmg/kgi.p.).After rats in each group got the corresponding treatment once per day for a week,open field test was used to detect locomotor activities and Quantitative Real-time PCR to detect the expression of miRNA-219 in prefrontal cortex of rats.Results: Compared with control group,the locomotor activity in MK-801 group increased and the expression of miRNA-219 decreased significantly(P〈0.01); and control group as well the locomotor activity in olanzapine+MK- 801 group decreased and the expression of miRNA-219 of increased significantly(P〈0.05 or P〈0.01). Conclusions: Olanzapine can inhibite the increase of locomotor activity and the decrease of miRNA-219 in frontal cortex of rats.
