摘  要:目的检测重组人γ-干扰素(recombination human interferon gamma,rh IFN-γ)对白血病K562细胞生长的抑制作用及其对CD123表达的影响.方法应用MTT法检测白血病K562细胞在不同浓度rh IFN-γ作用72 h的增殖活力以及用流式检测其CD123的表达.结果 5×10^4~10^8 U/L rh IFN-γ作用K562细胞72 h,细胞生长抑制率随剂量增大而增高,而2×10^5 U/L rh IFN-γ使其抑制率降低.rh IFN-γ2×10^5 U/L、10^7 U/L孵育K562细胞72 h,流式检测到CD123在K562细胞的表达量分别为(7.2±2.50)%和(21.6±1.17)%;10^7 U/L rh IFN-γ组与对照组(4.10±1.46)%比较,其差异有统计学意义(P〈0.05).结论 rh IFN-γ对白血病K562细胞生长有双向作用(抑制或促增殖),诱导CD123表达增加.Objective To investigate the inhibitory effect of rh IFN-γ on human chronic myeloid leukemia cell line K562, and the impaction on the expression of CD123.Methods MTT method was used to test cell relative viability with rh IFN-γ at different concentrations. The expression of CD123 on K562 cells was detected by flow cytometry. Results The relative inhibition of K562 cells proliferation was hampered when the cells were treated with rh IFN-γ for 72 h at the concentration of 5×10^4^10^8U/L,respectively. However, rh IFN-γ at 2×10^5U/L was benefit to K562 cells proliferation. After treatment with rh IFN-γ at 0, 2×10^5 U/L and 10^7 U/L,the percentages of CD123 expression on K562 cells were(4.10 ±1.46) %,(7.2 ±2.50) % as well as(21.6±1.17) %,respectively. Compared with the control group, 10^7 U/L rh IFN-γ significantly increased the expression of CD123 on K562 cells(P 〈0.05). Conclusion The effect of rh IFN-γ on the growth of K562 cells has two aspects(inhibition or proliferation),and it can increase the expression of CD123.
