广西壮族人群Nogo基因rs12464595和rs17046518与鼻咽癌的关系

摘　　要：目的:探讨Nogo基因多态性与广西壮族人群鼻咽癌的关系。方法:采用单碱基延伸PCR技术和DNA测序法,检测200例广西壮族鼻咽癌患者(鼻咽癌组)和200例壮族健康对照者(对照组)的Nogo基因rs12464595和rs17046518位点单核苷酸多态性,进而比较其相互间基因型和等位基因分布频率的差异,用SHEsis软件分析Nogo基因两位点的单倍型。结果:Nogo基因rs17046518位点的基因型和等位基因型频率在鼻咽癌组与对照组间分布差异无统计学意义(P>0.05)。但是rs12464595位点的T等位基因频率分布在两组间比较,差异有统计学意义(P=0.003)。单倍型分析的结果显示两者的CG和CA单倍型差异有统计学意义(P=0.045,P=0.002)。结论:在广西壮族人群中,携带Nogo基因位点rs12464595的T等位基因和CG单倍型以及CA单倍型可增加个体鼻咽癌的风险。Objective To investigate the relationship between genetic polymorphisms of Nogo gene and nasopharyngeal carcinoma in Zhuang ethnic group in Guangxi Province. Methods Nogo gene polymorphisms were analyzed between patients with nasopharyngeal carcinoma and healthy people as a control in Zhuang ethnic group in Gnangxi province using PCR-SBE. Their genotype and allele frequency distribution were compared between ease and control group. The frequencies of haplotypes were analyzed with SHEsis software between these two sites in different groups. Results There were no differences between the patients and controls in the genotype or allele frequencies of Nogo gene rs 17046518 site （P 〉 0.05）. But the frequency distribution of T allele was significantly different （P = 0.003） in the rs12464595 site. The result of haplotypes analysis showed that GA haplotype and CA haplotype were significantly different between cases and control （P = 0.045, P = 0.002）. Conclusion The CG haplotype, CA haplotype and T allele of Nogo gene rs12464595 site increase the risk of nasopharyngeal carcinoma among Zhuang ethnic group in Guangxi.
