摘 要:目的研究花椒提取物对致痤疮菌的体外抑菌活性,并初探其抑菌机制。方法以水蒸气蒸馏法提取花椒精油,热浸提法提取花椒水提物,采用纸片扩散法和96孔板微量肉汤稀释法,分别测定和比较2种花椒提取物对痤疮丙酸杆菌、表皮葡萄球菌、金黄色葡萄球菌的体外抑菌效果。选取抑菌效果好的提取物,以痤疮丙酸杆菌、金黄色葡萄球菌为受试对象,运用化学法探讨其对细菌胞壁通透性的影响。结果花椒精油对痤疮丙酸杆菌、表皮葡萄球菌、金黄色葡萄球菌的MIC分别为5mg/ml、16.7mg/ml、10mg/ml,花椒水提物对痤疮丙酸杆菌、表皮葡萄球菌、金黄色葡萄球菌的MIC均大于100mg/ml,花椒精油对3种致痤疮菌的抑菌效果优于水提物。MIC、2MIC浓度的花椒精油作用于痤疮丙酸杆菌、金黄色葡萄球菌可引起细菌细胞内K^+、Mg^(2+)、AKP等物质渗漏至胞外,抑制细菌生长。结论花椒精油对3种致痤疮菌均有明显的抑菌作用,抑菌效果优于花椒水提物,其抑菌机制可能是增加了细菌胞壁通透性,引起胞内物质外漏所致。Objective To study the in vitro antimicrobial effect of the extracts from Zanthoxylum bungeanum against acne - inducing bacterias and to analyze the related mechanism preliminarily. Methods We got the essential oil and the water extract using the ways of hydrodistillation and 80~C soaking extraction respectively. By K - B Disc Diffusion method and 96 - well plate 2 -fold dilution method, we measured and compared the in vitro inhibitory effects of the 2 kinds of Zanthoxylum bungeanum extracts against propionibacterium acnes, staphylococcus epidermidis and staphylococcus aureus, propionibacterium acnes and staphylococcus aureus were used as subjects to study the effect of the extract with better antimicrobial on membrane permeability by chemical methods. Results The MIC of the essential oil on propionibacterium aches, staphylococcus epidermidis and staphylococcus aureus were 5mg/ml, 16.7mg/ml, 10mg/ml respectively ,while the MIC of the water extract were all more than 100mg/ml,the essential oil had better antimicrobial effect against aene- inducing bacteria than the water extract. At the concentration of MIC and 2MIC, the essential oil could lead the leakage of intracellular K + , Mg2+ and AKP and inhibit the growth of bacteria. Conclusion The essential oil of Zanthoxylum bungeanum shows an obvious antimicrobial effect against 3 kinds of ache - inducing bacteria, which is better than the water extract, the mechanism may be related to the leakage of intracellular substances caused by membrane permeability.
