摘 要:目的了解丙戊酸对HL-60/HT细胞P27^(Kip1)的影响。方法建立白血病耐药细胞株HL-60/HT,MTT法检查丙戊酸作用前后细胞增殖情况及对Ara-c耐药性的改变,流式细胞仪检测HL-60、HL-60/HT细胞P27^(Kip1)、P170的表达。结果 VPA能增加HL-60、HL-60/HT细胞P27^(Kip1)的表达水平(P<0.05),而不影响P170的表达(P>0.05);Ara-c不能影响HL-60、HL-60/HT细胞的P27^(Kip1)、P170的表达(P>0.05);VPA与Ara-C联用能增加HL-60、HL-60/HT细胞P27^(Kip1)的表达水平(P<0.05),不影响P170的表达(P>0.05)。结论 HL-60/HT细胞P27^(Kip1)低于HL-60,丙戊酸能增加HL-60/HT细胞P27^(Kip1)的表达。Objective To understand the impact of valproie acid to expression level of P27^Kip1 and P170 of HL- 60/HT cells. Methods Leukemia resistant cell line HL-60/HT was established. MIT method was used to deteeeell proliferation before and after valproie acid and its changes to drug resistance of Ara-c. Flow eytometry was used to detect the expression of P27^Kip1 and PI70 of HL-60 and HL-60/HT ceils, Results VPA could increase the expression level of P27^Kip1 of HL-60 and HL-60/HT eells(P 〈 0.05). It did afloat the expression of P170(p 〉 0,05). Ara-e could not affect the expression level of P27^Kip1 of HL-60 and HL-60/HT eells(P 〈 0.05). VPA combined with Ara-C can increase the expression level of P27^Kip1 of HL-60 and HL-60/HT cells(P 〈 0.05). It did affect the expression of P170(P 〉 0.05). Conclusion P27^Kip1 of HL-60/HT cells is lower than HL-60 cells. Valproie acid can the expression of P27^Kip1 of HL-60/HT cells.
