摘  要:目的探讨低温保存对围生期间充质干细胞(P-MSCs)体外培养的生物学特性的影响。方法从脐带组织分离、培养P-MSCs,取第3代细胞梯度降温冻存后于液氮深低温保存6个月后复苏。比较低温保存前后PMSCs的增殖活性、细胞衰老情况、细胞表面标志物及向脂肪和成骨方向分化的潜能。未经低温保存的第3代P-MSCs作为对照。结果低温保存的P-MSCs增殖活性、衰老率与未冻存细胞相比差异无统计学意义(P>0.05)。低温保存的P-MSCs表达间充质干细胞的表面标志CD44、CD90和CD105,且能够向成骨和脂肪细胞分化。结论低温保存复苏后间充质干细胞仍保持良好的体外生长特性和多向分化的潜能。Objective To investigate the effects of cryopreservation on biological features of perinatal mesenchymal stem cells (P-MSCs). Methods P-MSCs were isolated from human umbilical cord and subcultured in vitro. The third-gen- eration P-MSCs were frozen with a controlled rate freezer and thawed 6 months after liquid nitrogen storage. The effect of eryopreservation on P-MSCs viability, senescence-associated [3-gal activity, cell surface markers and adipogenic and osteogenie differentiation property were evaluated. The third-generation P-MSCs without storage were taken as control. Results After cryopreservation, P-MSCs remained maintained a growth rate similar to the P-MSCs without storage, also had the similar senescence-associated ^-gal activity (P 〉 0.05). The P-!VISCs after cryopreservation also highly expressed CD44, CD90 and CDI05, and had the adipogenic and osteogeic differentiation potential. Conclusion P-MSCs maintain their biological characteristics and have the differentiation potential after cryopreservation and thawing.
