益气复生方对胃癌组织中STAT3及HIF-1表达的影响

摘　　要：目的观察益气复生方对胃癌组织中STAT3及HIF-1表达的影响。方法取2015年1月~2016年7月医院进行实验的BALB/c小鼠60只,建立BALB/c小鼠胃癌抑制瘤模型,根据处理方法不同分为对照1组、对照2组和观察组,每组20只。对照1组不进行任何处理,对照2组给予5-氟尿嘧啶注射液,观察组给予益气复生方。14 d后处死小鼠,采用免疫组织化学Envision法检测3组小鼠胃癌组织中STAT3的表达水平,免疫组织化学SP法检测3组小鼠胃癌组织中HIF-1的表达水平;对肿瘤组织进行HE染色,分析益气复生方对胃癌组织中STAT3及HIF-1表达水平的影响。结果观察组小鼠胃癌组织中STAT3及HIF-1的表达水平显著低于对照2组和对照1组(P<0.05);对照2组小鼠胃癌组织中STAT3及HIF-1的表达水平显著低于对照1组(P<0.05)。观察组小鼠STAT3及HIF-1的表达阳性率显著高于对照2组和对照1组(P<0.05);对照2组小鼠STAT3及HIF-1的表达阳性率显著高于对照1组(P<0.05)。免疫组化结果显示,3组小鼠均存在STAT3的阳性表达,表达量从低到高依次为观察组、对照2组、对照1组;观察组与对照2组HIF-1表达的差异无统计学意义(P>0.05),对照1组HIF-1表达相对较高。观察组肠黏膜绒毛轻度破坏,排列相对整齐,治疗后有所恢复;对照1组和对照2组肠黏膜绒毛完整性被破坏,部分绒毛坏死脱落、稀疏,排列紊乱。结论益气复生方可降低胃癌组织中STAT3的表达水平,抑制HIF-1的表达。Objective To observe the effects of Yiqi Fusheng Decoction on expressions of STAT3 and HIF-1 from gastric carcinoma. Methods From January, 2015 to July, 2016, 60 BALB/c mice were adopted to establish mouse gastric cancer BALB/c tumor-inhibiting model. According to the different treatment methods, they were divided into control group 1 （n=20）, control group 2 （n=20） and observation group （n=20）. The control group 1 received no treatment, while the control group 2 was given 5-Fluorouracil injection treatment. The ob- servation group was treated with Yiqi Fusheng Decoction. After treatment for 14d, the BALB/c mice were sacrificed. The STAT3 expression of gastric cancer tissues of BALB/c mice were detected by immunohistochemistry Envision method and HIF-1 expression were detected by immunohistochemistry SP method in the three groups. All tumor tissues got HE staining. Analyze the effects of Yiqi Fusheng Decoction on the expressions of STAT3 and HIF-1 in gastric carcinoma. Results The STAT3 and HIF-1 expression levels were lower in the gastric car- cinoma tissues of BALB/c mice in observation group than in the control group 1 and 2 （P 〈 0.05）. The expression levels of STAT3 and HIF- 1 were also lower in the control group 2 than in the control group 1 （P 〈 0.05）. The positive expression rate of STAT3 and HIF-1 were higher in the observation group than in the control group 1 and 2 （P〈0.05）, and they were also higher in the control group 2 than in the control group 1 （P〈0.05）. The immunohistochemical results showed that there was STAT3 positive expression in the three groups, but least in observa- tion group, and most in control group 1. However, HIF-1 expression showed no significant difference between the observation group and the control group 2 by immunohistochemical study （P〉O.05）, but was relatively higher in the control group 1.
