摘 要:目的探讨贝伐单抗对胃移植癌裸鼠血管内皮生长因子(VEGF)、白细胞介素-8(IL-8)、碱性成纤维细胞生长因子(bFGF)表达的影响。方法选择BALB/c裸鼠为研究对象,于颈背部皮下注射胃癌细胞株以建立胃移植癌裸鼠模型,随机分为空白对照组、低剂量贝伐单抗组和高剂量贝伐单抗组。低剂量组注射11 mg·kg^(-1)贝伐单抗,高剂量组注射22 mg·kg^(-1)贝伐单抗,空白对照组注射等体积生理盐水,比较三组裸鼠的肿瘤生长情况、血清以及肿瘤组织VEFG、IL-8、bFGF含量。结果治疗7 d、14 d、21 d后,贝伐单抗组肿瘤体积明显小于空白组,且高剂量贝伐单抗组明显小于低剂量贝伐单抗组(P<0.05)。治疗21d后,贝伐单抗组的肿瘤质量明显低于空白组,且高剂量贝伐单抗组明显低于低剂量贝伐单抗组(t=2.189~4.807,P<0.05);贝伐单抗组裸鼠血清与肿瘤组织中VEGF、IL-8、bFGF含量均明显低于空白组,且高剂量贝伐单抗组血清与肿瘤组织中VEGF、IL-8、bFGF含量明显低于低剂量贝伐单抗组(t=2.443~7.745,P<0.05)。结论贝伐单抗对胃移植癌裸鼠模型的肿瘤生长具有抑制作用,同时能够降低VEGF、IL-8、bFGF的表达。Objective To study the effects of bevacizumab on expressions of vascular endothelial growth factor (VEGF), Interleukin- 8(IL-8) and basic fibroblast growth factor (bFGF) in gastric carcinoma nude mice model. Methods BALB/c nude mice were collected as research subjects. The gastric carcinoma xenografts nude mice model were established by subcutaneous injection of gastric cancer cells, and randomly divided into blank control group, low dose bevacizumab group and high dose bevacizumab group. The blank control group were injected with saline, and low dose group with 11 mg-kg-l bevacizumab, while high dose group with 22 mg,kg-l bevacizumab. Then the tumor growth in nude mice, the VEGF, IL-8 and bFGF levels in both serum and tumor tissues were detected and compared between three groups. Results At 7 d, 14 d, 21 d after treatment, the tumor volume were significantly lower in bevacizumab groups than in the blank group, and it significantly lower in high dose bevacizumab group than in low dose bevacizumab group (P 〈 0.05). At 21 d after treatment, the tumor weight were significantly lower in bevacizumab groups than in blank group, and also were lower in high dose bevacizumab group than that in low dose bevacizumab group (t=2.189 ~ 4.807, P 〈 0.05). The VEGF, IL-8 and bFGF levels in serum and tumor tissues in bevacizumab groups were significantly lower than in the blank group, and also significantly lower in high dose bevacizumab group than in low dose bevacizumab group 0=2.443 ~ 7.745, P 〈 0.05). Conclusion Bevacizumab had inhibiting effect on tumor growth of gastric carcinoma nude mice model, and meanwhile reduced the expression levels of VEGF, IL-8 and bFGF.
