摘  要:目的应用超声微泡介导碱性成纤维生长因子(bFGF)基因进行受损面神经(大鼠模型)修复,研究其可行性及有效性。方法将40只SD大鼠制作面神经损伤模型,并分成4组,每组10只;A组:bFGF+超声+微泡组;B组:bFGF+微泡组;C组:bFGF+超声组;D组:单纯手术组(PBS)。于bFGF基因转染后第1、10、20、28天,观察各组大鼠一般状况,检测损伤面神经传导速度、潜伏期及动作电位波幅。提取损伤处面神经组织后采用RT-PCR检测mRNA的表达,Western blot检测bFGF蛋白表达。结果转染后第20天,A组大鼠术侧有少量胡须可观察到细微摆动;转染后第28天,A组大鼠一般状况较B、C、D 3组恢复得更好。A组面神经损伤后修复的神经电生理表现优于B、C、D 3组;A组bFGF mRNA和蛋白表达量明显高于B、C、D 3组(P<0.05)。结论超声微泡介导bFGF有助于面神经损伤的修复。Objective To apply the ultrasound microbubble to mediate basic fibroblast growth factor(bFGF) for conducting the injuried facial nerve(rat model) repair and to investigate its feasibility and efficiency. Methods After establishing the models of facial nerve injury,40 SD rats were divided into 4 groups, 10 eases in each group:group A, bFGF + uitrasoundq-microbubble(bFGF + MB/US), group B, bFGF and mierobuble( bFGF+ MB), group C, bFGF and ultrasound (bFGF +US) and group D, simple opera- tion(PBS). The general status of rats on 1,10,20,28 d after bFGF gene transfection was observed. The nerve conduction velocity (NCV) ,incubation period and amplitude of facial nerve action potential were measured. After taking the facial nerve tissue in inju- tied site,mRNA expression was detected by RT-PCR. Western blot was used to detect the bFGF protein expression. Results On 20 d after transfectlon, small swing of a small quantity of beard in the operation site of the group A could be observed;on 28 d after transfeetion,the general slatws of reeavely in rats m the grouprA was better than that in the group B,C and D. The nerve electro- physiology manifestations after facial nerve repair in the group A were superior to the group B,C and D; the amount of hFGF mR- NA and protein pxpression in the group A was significantly higher than that in the group B,C and D. Conclusion Ultrasound mi- erobubble mediated bFGF is conducive to the repair of facial nerve injury.
