摘 要:目的探讨中期因子(MK)、血管内皮生长因子(VEGF)和基质金属蛋白酶-2(MMP-2)在结直肠癌组织中的表达及临床意义。方法选取2013年2月至2016年2月89例结直肠癌患者癌组织,同时选取癌旁组织作为对照,采用免疫组织化学检测MK、VEGF和MMP-2表达。数据分析采用SPSS 19.0软件,两种组织中MK、VEGF及MMP-2阳性表达率以及与临床特征关系比较使用χ2检验,三者的表达相关性采用Spearman秩相关分析。P<0.05表示差异具有统计学意义。结果结直肠癌组织MK、VEGF及MMP-2阳性表达率分别为80.9%、83.2%和89.9%,明显高于癌旁组织(P<0.05);Duke分期C-D期患者MK、VEGF及MMP-2阳性表达率分别为93.4%、93.4%和96.7%,明显高于A-B期患者(P<0.05);有淋巴结转移患者MK、VEGF及MMP-2阳性表达率分别为93.6%、93.6%和97.9%,明显高于无淋巴结转移患者(P<0.05);MK、VEGF及MMP-2阳性表达与结直肠癌患者性别、年龄、肿瘤大小、部位及组织分化无关(P>0.05);结直肠癌组织MK表达与MMP-2和VEGF表达呈正相关(r=0.774和0.406,P<0.05)。结论 MK、VEGF及MMP-2在结直肠癌组织中的高表达,与Dukes分期和淋巴结转移有一定关系。Objective To investigate expression and clinical significance of midkine(MK),vascular endothelial growth factor(VEGF)and matrix metalloproteinase-2(MMP-2)in colorectal cancer.Methods From February 2013 to February 2016,89 patients with colorectal cancer were enrolled,of whom both cancer tissues and carcinoma adjacent tissues were harvested and the expression of MK,VEGF and MMP-2 were detected by using immunohistochemistry staining,Statistical analysis were performed by using SPSS 19.0 software.Count data,such as expression rate of MK,VEGF and MMP-2 and clinical features,were expressed as%,and were examined by using x2 test.The correlation of the three expressions was analyzed by Spearman rank correlation analysis.A P value 〈0.05 was considered as statistically significant difference.Results The positive expression rates of MK,VEGF and MMP-2 in colorectal carcinoma were 80.9%,83.2%and 89.9%respectively,which were significantly higher than those in carcinoma adjacent tissue(P〈0.05).The positive expression rates of MK,VEGF and MMP-2 in Duke stage C-D patients were 93.4%,93.4%and 96.7%respectively,which were significantly higher than those in patients with Duke stage A-B(P〈0.05).The positive expression rates of MK,VEGF and MMP-2 in patients with lymph node metastasis were 93.6%,93.6%and 97.9%respectively,which were significantly higher than those without lymph node metastasis(P〈0.05).The positive expression of MK,VEGF and MMP-2 was not related to sex,age,tumor size,location and tissue differentiation of colorectal cancer patients(P〉0.05).The expression of MK in colorectal carcinoma was positively correlated with MMP-2 and VEGF expression(rs=0.774 and 0.406,P〈0.05).Conclusion MK,VEGF and MMP、2 are highly expressed in colorectal cancer,and have a certain relationship with Dukes staging and lymph node metastasis.
