摘  要:目的观察普罗布考对血管性痴呆(VD)大鼠的认知功能及海马区JNK、p-JNK表达的影响。方法将30只雄性SD大鼠随机分为假手术组、模型组、普罗布考组,采用永久性结扎双侧颈总动脉的方法制作VD大鼠模型,灌胃给予普罗布考8w后通过Morris水迷宫观察大鼠的学习记忆能力,采用流式细胞术评估大鼠海马神经元凋亡率,免疫组织化学法和蛋白印迹(Western blot)法检测大鼠海马c-Jun氨基末端激酶(JNK)、磷酸化的c-Jun氨基末端激酶(p-JNK)的表达。结果与假手术组相比,模型组大鼠学习记忆能力明显减弱,大鼠海马细胞凋亡率显著增高,海马区p-JNK蛋白的表达水平增高。应用普罗布考后大鼠学习记忆能力明显提高,海马细胞凋亡率显著降低,海马区p-JNK蛋白的表达水平降低。结论普罗布考可以改善VD大鼠的学习记忆能力,其机制可能与其抑制JNK信号通路的异常激活有关。Objective To observe the effect of probucol on vascular dementia ( VD ) rats in cognitive function and expression of JNK and p-JNK protein. Methods Thirty male SD rats were randomly divided to shame operated group, model group and probucol group. The VD rat model was established with the method of permanent ligation of bilateral common carotid artery. After intragastric administration of probucol for 8 weeks, Morris water maze experiment was used to evaluate learning and memory ability of rats, and flow cytometry was used to assess the apoptotic rate of hippoeampus nerve cells. Meanwhile, the expression levels of JNK and p-JNK protein in the hippocampus were examined by immunohistochemical staining and western blot. Results Compared with shame operated group, learning and memory ability of rats in model group decreased significantly. On the other hand, apoptosis rate of hippocampus neurons and expression of p-JNK protein increased significantly. Compared with model group, learning and memory ability of rats in probucol group improved significantly. Apoptosis rate and expression level of p-JNK protein in the hippocampus of rats in probucol group decreased significantly. Conclusion The probucol could significantly improve the learning and memory ability of rats with VD. The protective effect may be related to the down regulation of JNK signal pathway.
