摘  要:目的探究Fut8基因敲除对ICR小鼠肠道菌群结构的影响。方法分别在Fut8^(+/+)和Fut8^(-/-)小鼠出生后不同时间点检测体重,采集粪便样本,采用PCR-变性梯度凝胶电泳(PCR-DGGE)的方法检测小鼠粪便中菌群的组成;利用高效液相色谱法(HPLC)测定Fut8^(+/+)和Fut8^(-/-)小鼠Fut8酶活。结果 Fut8^(-/-)小鼠与Fut8^(+/+)小鼠相比生长缓慢(P<0.05);完全丧失核心岩藻糖基化活性;肠道菌群结构不同,其中拟杆菌门微生物存在显著差异。结论核心岩藻糖基转移酶Fut8基因敲除对小鼠肠道菌群结构有显著影响。Objective To determine the effect of Fur8 gene knockout on the structure of intestinal microbiota in ICR mice. Methods The body weights were measured at different time points after Fur8+/+ and Fut8 / mice were born. Fecal samples were collected at the same time to detect the intestinal microbiota by using PCR-Denaturing Gradient Gel Electrophoresis (PCR-DGGE) . The enzyme activity of Fut8 was assayed by using High Performance Liquid Chromatography (HPLC) . Results The Fur8-/- mice grew slowly com- pared with Fut8+/+ mice (P^0.05), and the fucosyltransferase activity was abolished. Compared with the controls, the intestinal microbiota of Fut8-/ mice showed a different pattern, among which the bacteria that belong to the phylum of Bacteroidetes changed significantly. Conclusion Core fucosyltransferase (FurS) gene knockout had a significant effect on the structure of intestinal microbiota in mice.
