摘  要:目的 探讨MAT2B基因在肝癌组织的表达及其在肝癌形成和发展中的作用。 方法 收集8例的手术切除肝癌组织标本分别由反转录-聚合酶链反应(RT-PCR)和62例病理组织切片免疫组织化学测定肝癌组织中MAT2B的表达,10例外伤正常肝组织作为对照。对癌旁及肝癌组织MAT2B表达阳性患者进行随访。结果 MAT2B基因在正常肝组织中没有表达,肝癌组织中MAT2B的表达阳性率为80.65%,显著高于癌旁组织的62.90%。RT-PCR检测肝癌组织、癌旁组织的表达分别为1.05±0.12、0.48±0.06和0;MAT2B的表达与肝癌的大小以及是否合并有肝硬化明显相关,与患者年龄、性别、分化程度、分期无明显相关。癌旁组织MAT2B阳性者较癌旁组织MAT2B阴性者、肝癌组织MAT2B阳性者生存时间明显缩短。 结论 MAT2B基因的表达与肝癌的形成及预后有关。Objective To investigate the expression of MAT2B gene and its significance in the tumorigeness and progress of in hepatic cancer.Methods 10 fresh tissues of hepatic carcinoma from radical resection of hepatoma were collected to be examined with reverse transcription-polymerase chain reaction (RT-PCR) and 62 pathological histological section be examined with Immunohisto-chemistry staining, 10 normal liver tissues were used as control group. All patients followed up monthly for decision of plan in next step.Results The expression of MAT2B gene in hepatic carcinoma tissues (80.65%) was higher than that in pericarcinomatous tissues (62.90%). The relative value of MAT2B was determined by RT-PCR was 1.05±0.12、0.48±0.06 and 0 respectively. the expression of MAT2B gene was relative with size of tumor and whether it company with hepatic cirrhosis or not. On the other hand the expression of MAT2B gene was not relative with age sex and degree of differentiation of tumor Patients of para-carcinoma with positive expression were less survival time than that of negative expression. Conclusion The expression of MAT2B gene is relative with tumorigeness and progress of in hepatic cancer. It can be used as early diagnosis index of hepatic carcinoma.
